Indications
Atorvastatin is recommended as a dietary supplement for patients with primary hypercholesterolemia, heterozygous familial and non-familial hypercholesterolemia, and mixed dyslipidemia in order to lower elevated levels of total cholesterol, LDL cholesterol, apolipoprotein B, and triglycerides (Fredrickson types IIa and IIb) Those with primary dysbetalipoproteinemia (Fredrickson type III) and elevated serum TG levels (Fredrickson type IV) who do not respond well to dietary changes. As a supplement to other lipid-lowering therapies (such as LDL apheresis) or in the absence of such therapies, homozygous familial hypercholesterolemia.
Pharmacology
HMG-CoA reductase may be specifically inhibited by atorvastatin. The rate-limiting protein for the conversion of HMG-CoA to mevalonate, a precursor to sterols including cholesterol, is this chemical. By blocking HMG-CoA reductase and cholesterol union in the liver and increasing the number of hepatic LDL receptors on the cell surface for better uptake and catabolism of LDL, atorvastatin lowers plasma cholesterol and lipoprotein levels.
