How does Orsema 0.50mg Subcutaneous Injection work?

• Acts on brain areas suppressing appetite and inducing a feeling of fullness.
• When combined with diet and exercise, it leads to significant weight loss.

Side Effects of Orsema 0.50mg Subcutaneous Injection

• Generally mild.
• Common side effects include dizziness, fatigue, gastrointestinal issues (diarrhea, constipation, gassiness), headache, and stomach issues (nausea, vomiting, pain, distension).

Quick Suggestions:

• Take Semaglutide as advised by your physician. It should be taken on an empty stomach. After taking Semaglutide wait at least 30 minutes before having your first meal or drink of the day or taking other oral medicines. Do not stop using this medicine without telling your doctor. Otherwise, your blood sugar levels may raise
• Stay hydrated by drinking plenty of water throughout the day
• Attend regular health checkups while taking this medicine
• Inform your healthcare provider about all the medications, supplements, and herbal products you are using, as Semaglutide can interact with certain drugs
• Semaglutide is not recommended for use in pregnant or breastfeeding women

Indication

Type 2 Diabetes Mellitus (T2DM): Adjunct to diet and exercise for improving glycemic control in adults.

Pharmacology

GLP-1 Agonist:

• Enhances glucose-dependent insulin secretion.
• Suppresses inappropriately elevated glucagon secretion.
• Slows gastric emptying.

Dosage & Administration:

Adult Dose:

Subcutaneous Administration:

• 0.25 mg SC: Once weekly for 4 weeks initially.
• Dose Escalation:
  • Increase to 0.5 mg SC once weekly.
  • If glycemic control is not achieved, can be further increased to 1 mg SC once weekly.
• The initial 0.25-mg dose is for treatment initiation and is not effective for glycemic control.

Child Dose:

• <18 years: Safety and efficacy not established.

Renal Dose:

• Renal impairment: No dosage adjustment is required.

Administration:

• SC Administration:
  • Administer to the abdomen, thigh, or upper arm.
  • Once weekly, on the same day each week, at any time of the day, with or without meals.
  • Change the injection site weekly when injecting in the same body region.

Interaction

• Insulin secretagogues or insulin may increase the risk of hypoglycemia.
• Caution with oral medications due to delayed gastric emptying.

Contraindications

Avoid individuals with:

• History of medullary thyroid cancer.
• History of gallbladder disease.
• History of pancreatitis.
• Multiple endocrine neoplasia syndrome type 2 (MEN2).

Pregnancy & Lactation

• Pregnancy:
  • Limited data; potential risks to the fetus.
  • Use during pregnancy if benefits justify potential risks.
  • Discontinue treatment at least 2 months before a planned pregnancy.
• Lactation:
  • No data on the presence of semaglutide in human milk.
  • Detected in rat milk at lower levels than in maternal plasma.

Precautions & Warnings

• Based on animal studies, semaglutide may cause thyroid C-cell tumors, including MTC.
• Monitor for signs and symptoms of pancreatitis (persistent severe abdominal pain, possibly radiating to the back, with or without vomiting).
• Patients with a history of diabetic retinopathy may have an increased risk for complications.
• Semaglutide pens must not be shared between patients.
• Monitor renal function in patients reporting severe adverse gastrointestinal reactions.

Storage Conditions

• Store your Orsema in the refrigerator at 2°C to 8°C (36°F to 46°F).
• After the first use of Orsema, the pen can be stored for 56 days in a refrigerator (2°C to 8°C).
• Do not freeze Orsema. Do not use it if it has been frozen.
• Unused Orsema cartridges may be used until the expiration date printed on the label, as long as they are kept in the refrigerator (2°C to 8°C).
• Keep Orsema away from heat.
• Store Orsema away from direct light.
• Keep the pen cap on when Orsema is not in use.
