Dienogest acts as a progesterone receptor agonist with moderate receptor affinity but a strong progestational effect on endometrial tissue. Prolonged administration leads to endometrial atrophy.
Its therapeutic effects include:
Continuous use creates a hyperprogestogenic and moderately hypoestrogenic environment, resulting in decidualization and suppression of both eutopic and ectopic endometrium. Dienogest also exhibits anti-androgenic activity, which may improve androgen-related symptoms such as acne and hirsutism.

Endosis is indicated for the treatment of endometriosis. It belongs to the class of medicines known as progestins, which counteract the effects of estrogen on estrogen-sensitive tissues such as the endometrium. By suppressing estrogen-induced growth of endometrial tissue, Endosis helps reduce inflammation and relieve pelvic pain associated with endometriosis.
It is unknown if Dienogest interacts with alcohol.

Adverse effects are more common during the initial months of therapy and generally decrease with continued use.
Dienogest is primarily metabolized by the CYP3A4 enzyme system. Concomitant use with CYP3A4 inhibitors may increase plasma levels and the risk of adverse effects.
Notable interacting drugs include:
Clinical studies indicate no significant risk of acute toxicity following overdose. Doses of 20–30 mg/day over 24 weeks were well tolerated. No specific antidote is required; supportive care is sufficient.
Pregnancy: Endosis should not be used during pregnancy, as treatment of endometriosis is not required during this period.
Lactation: Use is not recommended during breastfeeding due to potential excretion in breast milk. A risk-benefit decision should be made regarding the continuation of breastfeeding or therapy.
