✔️ How does Zoleta 2.5 work?

Zoleta 2.5 acts by blocking the actions of an enzyme called aromatase in the body which is involved in the production of estrogen. This action suppresses the release of excessive estrogen in the body which triggers breast cancer.

• Avoid smoking and alcohol consumption.

• Do not take Letrozole if you are pregnant or breastfeeding
• Avoid fast food, fried food, processed meats, refined carbs, and added sugar.

✔️ Indication of Zoleta 2.5

1. Adjuvant Therapy:
   • For postmenopausal women with hormone receptor-positive early breast cancer.
   • For postmenopausal women with early breast cancer (positive or unknown estrogen/progesterone receptor status) after 5 years of adjuvant tamoxifen therapy (extended adjuvant therapy).
2. First-Line Treatment:
   • For hormone-dependent advanced breast cancer in postmenopausal women.
3. Advanced Breast Cancer:
   • For women with natural or induced postmenopausal status previously treated with anti-estrogens.
4. Pre-Operative Therapy:
   • For localized hormone receptor-positive breast cancer in postmenopausal women to enable breast-conserving surgery when initially deemed ineligible.

✔️ Pharmacology

Mechanism of Action:

• Letrozole is a nonsteroidal aromatase inhibitor that reduces estrogen biosynthesis by inhibiting the aromatase enzyme.
• It prevents the conversion of adrenal androgens (androstenedione and testosterone) to estrogens (estrone and estradiol), thereby reducing the growth stimulus for estrogen-dependent breast cancer cells.

Effects on Estrogen Levels:

• Suppresses serum estrone and estradiol by 75-95%, with maximum suppression achieved within 48-78 hours.

Pharmacokinetics:

• Absorption: Rapidly absorbed with almost complete bioavailability (99.9%).
• Food Impact: Food slightly slows absorption but does not affect its extent.
• Protein Binding: Approximately 60%, primarily to albumin.
• Metabolism: Metabolized slowly to an inactive carbinol metabolite via CYP3A4 and CYP2A6 enzymes.
• Half-life: Approximately 2 days.
• Elimination: Steady-state levels achieved in 2-6 weeks with daily dosing.

✔️ Dosage & Administration of Zoleta 2.5

• Recommended Dose: 2.5 mg once daily.
• Duration:
  • Adjuvant and extended adjuvant therapy: Up to 5 years or until tumor relapse.
  • Metastatic disease: Continue until tumor progression.
• Elderly Patients: No dose adjustment is required.
• Hepatic/Renal Impairment: No adjustment is needed unless severe hepatic impairment (monitor closely).
• Children: Not applicable.

Take Zoleta 2.5 with or without food. Swallow Zoleta 2.5 as a whole with a glass of water; do not chew or break it. Take Zoleta 2.5 at the same time every day.

Drug-Drug Interactions:

• Antidepressants: (e.g., citalopram, escitalopram).
• Anticancer drugs: (e.g., tamoxifen).

Drug-Food Interactions:

• No significant interactions have been reported.

Drug-Disease Interactions:

• Inform your doctor if you have:
  • High cholesterol levels.
  • Osteoporosis or a history of bone disorders.
  • Liver disease.

• Pregnancy: Contraindicated due to risks of teratogenicity and fetal toxicity observed in animal studies.

• Avoid alcohol as it may increase drowsiness or dizziness.
