Indications of Lopirel Tablet

Lopirel 75 is a blood thinner or antiplatelet medication that aids in preventing the development of harmful blood clots in your blood vessels. This reduces your risk of having a heart attack or stroke. A lot of people take this medication to protect their hearts. People with a high risk of developing heart disease are given Lopirel 75. This includes those who have peripheral vascular disease (problems with blood circulation brought on by narrowing of blood vessels), have recently experienced a heart attack or stroke, have irregular heartbeats, or have had certain heart procedures like stenting. It is also used in conjunction with other medications to treat certain types of heart-related chest pain and heart attacks (unstable angina)

Pharmacology of Lopirel Tablet

A prodrug used to treat blood clots is clopidogrel. The P2Y12 class of ADP receptors on platelets serves as an irreversible binding site for its active metabolite, which inhibits platelet activation and aggregation. A dose-dependent suppression of platelet aggregation can be observed two hours after a single oral dose. Repeated 75 mg doses on day one reduce ADP-induced platelet aggregation, and between days three and seven, inhibition reaches a stable state.

Aspirin stops the production of thromboxane A2, a powerful inducer of platelet aggregation and vasoconstriction, by permanently inhibiting platelet cyclooxygenase.

Dosage & Administration of Lopirel Tablet

• Acute Coronary Syndrome: Start clopidogrel with a single oral loading dose of 300 mg (4 tablets) in patients who require an antiplatelet effect within hours. Then, continue with 75 mg once daily. It will take several days longer to establish an antiplatelet effect if you start it without a loading dose.
• Patients with established peripheral arterial disease, recent MI, or recent stroke should take 75 mg orally once daily without a loading dose.
• It can be administered orally with or without food.

Interactions of Lopirel Tablet

The risk of bleeding is raised by oral anticoagulants, NSAIDs, Metamizole, SSRIs, and CYP2C19 inhibitors, among other medications. It interacts with tonofovir, valproic acid, varicella vaccine, acetazolamide, and nicorandil.

Contraindications

If you have any of the following conditions, you shouldn't take this combination: hypersensitivity to the drug or any of the ingredients in the product. Examples of current pathological bleeding include cerebral hemorrhage and peptic ulcers.

Side Effects of Lopirel Tablet

This combination is generally well tolerated.

Pregnancy & Lactation

In pregnant women, there are no sufficient and well-controlled trials. It should only be taken if absolutely necessary throughout the first and second trimesters of pregnancy. During the third trimester of pregnancy, it is not recommended. Clopidogrel is not known to be excreted in human breast milk, however, it is known to be excreted in human milk. During breastfeeding, this drug should be avoided.

Precautions & Warnings

• TTP (thrombotic thrombocytopenic purpura) has only occasionally been reported as an adverse reaction to this mixture.
• People are afflicted with Reye's syndrome. Reye's syndrome can appear in people who have chicken pox, the flu, or flu-like symptoms. Patients receiving this combination or those with a history of thienopyridine hypersensitivity have experienced hematologic, rash, or angioedema reactions.
• Your chance of bleeding rises as a result. Use caution when using sharp objects, shaving, or cutting your fingernails or toenails.
• Before having any surgery, let the surgeon know you are taking Lopirel Plus.
