Omeprazole, the active ingredient, inhibits gastric acid secretion by blocking the H+/K+ ATPase enzyme system in gastric parietal cells. It exhibits an antisecretory effect within one hour of administration, with a maximum effect within two hours, lasting up to 72 hours. Upon discontinuation, secretory activity gradually returns over 3 to 5 days.
Dosage varies depending on the condition treated. For example, for benign gastric and duodenal ulcers, the typical dose is 20 mg once daily for 4 weeks in duodenal ulceration and 8 weeks in gastric ulceration. Maintenance doses may differ for different conditions
Seclogen may interact with several drugs, affecting their absorption or metabolism. For example, it may decrease the absorption of ketoconazole and delay the elimination of diazepam, phenytoin, and warfarin.
Omeprazole is contraindicated in patients with known hypersensitivity to it. It should be used cautiously in patients with suspected gastric ulcers, as treatment may mask symptoms and delay diagnosis of malignancy.
Seclogen can be used during pregnancy, but breastfeeding should be discontinued if its use is deemed essential.
Avoid concomitant use with clopidogrel, as it may reduce the pharmacological activity of clopidogrel. Long-term use may lead to atrophic gastritis, and caution should be exercised when co-administering with methotrexate.
