✔️ Pharmacology

Esomeprazole, a proton pump inhibitor (PPI), suppresses gastric acid secretion by inhibiting the H⁺/K⁺-K-ATPase enzyme system in gastric parietal cells. This action blocks the final step in acid production, providing effective relief in acid-related disorders and promoting ulcer healing.

✔️ Dosage & Administration

✔️ Administration:

Swallow the tablet whole with water. Do not crush or chew. If swallowing is difficult, disperse in half a glass of non-carbonated water, stir, and drink within 30 minutes. Do not chew the pellets.

✔️ Interaction

• Metabolism: Primarily via CYP2C19 and CYP3A4.
• May reduce clearance of diazepam (a CYP2C19 substrate).
• May alter absorption of pH-dependent drugs (e.g., ketoconazole, digoxin, iron salts).
• Co-administration with clarithromycin and amoxicillin increases plasma levels of Esomeprazole and its metabolites.
• No significant interactions reported with phenytoin, warfarin, or oral contraceptives.

Known hypersensitivity to Esomeprazole or any component of the formulation.

✔️ Common adverse reactions may include:

• Headache
• Nausea
• Abdominal pain
• Diarrhea or constipation
• Flatulence
• Dry mouth
• Long-term therapy may slightly increase the risk of bone fractures (hip, wrist, spine).
