Pharmacology

Beklo 5reduces the monosynaptic and polysynaptic reflexes in the spinal cord by activating GABAB receptors, which inhibit the release of glutamate and aspartate. The drug may also potentially depress the central nervous system through its action at extraspinal sites. Additionally, Baclofen exhibits antinociceptive properties.

Dosage & Administration of Beklo 5

Adult & children over 10 years: 5 mg 3 times daily, preferably with or after food, gradually increased; max. 100 mg daily.

Children <10 years: Treatment is usually started with 2.5 mg (2.5 ml) given 4 times daily and then raised according to requirement. Daily maintenance dose-

• 12 month-2 years: 10-20 mg (10-20 ml)
• 2 years-6 years: 20-30 mg (20-30 ml)
• 6 years-10 years: 30-60 mg (30-60 ml)

Interaction of Beklo 5

• Taking Beklo 5 concurrently with medications that affect the central nervous system, alcohol, or artificial sedatives may result in increased sedation and a higher risk of respiratory depression. Combining Baclofen with antihypertensive medication may prolong the drop in blood pressure, and appropriate dosage management is necessary.
• Concurrent use of Beklo 5 and tricyclic antidepressants may amplify the drug's pharmacological effects resulting in pronounced muscle hypotonia.
• Parkinson's disease patients taking Baclofen, levodopa, and carbidopa have reported mental confusion, hallucinations, headaches, nausea, and agitation.
• Combining Baclofen with MAO inhibitors may have more profound CNS depressive effects, and appropriate dosage adjustments of one or both agents should be made with caution.
• Care should be taken when using Beklo 5 and magnesium sulfate or other neuromuscular blocking drugs together as they may interact synergistically.

Contraindications

Individuals who have a severe sensitivity to any of Baclofen's components should avoid using the product.

Pregnancy & Lactation

Beklo 5 falls under pregnancy category B3, and its safe usage during pregnancy has not yet been established. The drug can cross the placental barrier, and its administration to pregnant women should be based on the physician's assessment of whether the potential benefits outweigh the possible risks.

Beklo 5 is excreted in breast milk, but available evidence suggests that the quantities are minimal, and no adverse effects on the infant are anticipated.

Precautions & Warnings

• Patients with a history of mania, schizophrenia, depression, hyper-depressive states, or confessional moods should be closely monitored when treated with Baclofen as these conditions may worsen. Similarly, individuals with impaired renal function or undergoing continuous hemodialysis should be administered lower dosages of around 5 mg per day.
• Baclofen withdrawal or overdose may result in seizures and lower writhing thresholds. Therefore, it is crucial to monitor patients in such cases.
• Baclofen may be administered to individuals with epilepsy and muscle spasticity, provided they are under proper monitoring and ongoing effective anticonvulsive treatment.
• Patients with underlying peptic ulcers, cerebrovascular diseases, or hepatic, renal, or respiratory failure should exercise caution while using Beklo 5. Additionally, regular laboratory tests must be conducted to ensure that medication-induced changes have not occurred in patients with hepatic disorders or diabetes mellitus.
