Dasacent (Dasatinib)
- Treatment of adult patients with:
- Newly diagnosed Philadelphia chromosome-positive (Ph+) chronic myeloid leukemia (CML) in chronic phase.
- Chronic, accelerated, or myeloid/lymphoid blast phase Ph+ CML with resistance or intolerance to previous therapy, including Imatinib.
- Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL) with resistance or intolerance to previous therapy.
- Treatment of pediatric patients (1 year and older) with Ph+ CML in the chronic phase and newly diagnosed Ph+ ALL in combination with chemotherapy.

- Dasatinib inhibits several kinases, including BCR-ABL, SRC family, c-KIT, EPHA2, and PDGFRβ. It helps overcome resistance to Imatinib in leukemic cells by targeting mutations and alternative signaling pathways.
- Absorption: Dasatinib reaches maximum plasma concentrations within 0.5–6 hours after oral administration.
- Distribution: Highly protein-bound (96%) with an apparent volume of distribution of 2505 L.
- Elimination: Half-life is 3-5 hours, and 85% of the dose is excreted in the feces.
- Metabolism: Primarily metabolized by CYP3A4, with the active metabolite accounting for 5% of exposure.

✔️ Side Effects: Dasacent 50
- Myelosuppression, bleeding, fluid retention, cardiovascular events, pulmonary hypertension, QT prolongation, severe dermatologic reactions, tumor lysis syndrome.
✔️ Dosage & Administration: Dasacent 50
- Adults:
- Chronic phase CML: 100 mg orally once daily.
- Accelerated phase CML, blast phase CML, or Ph+ ALL: 140 mg orally once daily.
- Do not crush, cut, or chew tablets.
- Pediatric Patients (1 year and older):
- Doses based on body weight, ranging from 40 mg (10-20 kg) to 100 mg (≥45 kg) once daily.
- Monitor growth and development in children.
✔️ Drug Interactions:
- CYP3A4 Inhibitors/Inducers: Avoid co-administration with strong CYP3A inhibitors or inducers. Adjust Dasacent dosage if necessary.
- Gastric Acid Reducing Agents: Can reduce Dasatinib concentrations. Avoid H2 antagonists and proton pump inhibitors; use antacids with caution.

- Hypersensitivity to Dasatinib or any component of the formulation.
✔️ Pregnancy & Lactation:
- Dasatinib may harm the fetus. Avoid pregnancy during treatment and for 30 days after.
- Breastfeeding is not recommended during treatment and for 2 weeks after.
✔️ Precautions & Warnings:
- Myelosuppression: Monitor blood counts, especially in advanced phases.
- Bleeding Risks: Increased risk of hemorrhage, especially with anticoagulants or platelet inhibitors.
- Fluid Retention: Manage fluid retention with diuretics or steroids if necessary.
- Cardiovascular Issues & PAH: Monitor for cardiac dysfunction and signs of pulmonary arterial hypertension.
- QT Prolongation & Dermatologic Reactions: Monitor for prolonged QT and severe skin reactions.
- Tumor Lysis Syndrome: Ensure proper hydration and electrolyte balance, particularly in high tumor burden cases.
