The biologically active form of Vitamin D3, Calcitriol, exerts its effects by binding to Vitamin D receptors (VDRs) present in various body tissues.
Being fat-soluble, Vitamin D3 has an approximate half-life of 50 days. It is absorbed in the small intestine, binds to alpha-globulins, and is transported to the liver, where it is converted into 25-hydroxy Vitamin D3 (Calcidiol).
A subsequent hydroxylation in the kidneys produces 1,25-dihydroxy Vitamin D3 (Calcitriol), the active metabolite responsible for promoting calcium absorption.
Unmetabolized Vitamin D3 is stored mainly in fat and muscle tissues and excreted through urine and feces.

Ortho-D3 plays a vital role in promoting effective calcium and phosphate absorption, ensuring healthy bone and teeth formation, and preventing rickets, osteomalacia, and osteoporosis.

✔️ Dosage & Administration | Ortho D3
Capsule (Adults):
- Treatment of deficiency: 40,000 IU once weekly for 7 weeks.
Maintenance: 1,400–2,000 IU daily.
Serum 25-hydroxy Vitamin D levels should be rechecked after 3–4 months. - Prevention of deficiency: 20,000 IU every 4 weeks (higher doses may be required in some cases).
Adjunct in osteoporosis therapy: 20,000 IU once monthly.
Capsule (Children 12–18 years):
Treatment: 20,000 IU every 2 weeks for 6 weeks.
Prevention: 20,000 IU every 6 weeks.
Film-Coated Tablets:
1,000 IU (1–2 tablets) daily with food or within 1 hour after meals.
Oroflash / Chewable Tablets:
1,000–2,000 IU daily with food or within 1 hour after meals. Chew before swallowing.

- Phenytoin, barbiturates: Reduce Vitamin D efficacy by increasing metabolism.
- Glucocorticoids: Decrease Vitamin D effect.
- Cardiac glycosides (e.g., digoxin): Risk of toxicity; ECG and calcium monitoring recommended.
- Ion exchange resins/laxatives (e.g., cholestyramine, paraffin oil): May reduce absorption.
- Actinomycin, imidazole antifungals: Inhibit the conversion of Vitamin D3 to its active form.
