✔️ Pharmacology:

The active form of Vitamin D3, Calcitriol, binds to Vitamin D receptors (VDRs) distributed in body tissues. Vitamin D3 is absorbed in the small intestine, metabolized to 25-hydroxy Vitamin D3 (Calcidiol) in the liver, and further hydroxylated to 1,25-dihydroxy Vitamin D3 (Calcitriol) in the kidney. This metabolite increases calcium absorption. Non-metabolized Vitamin D3 is stored in tissues. It is eliminated via feces and urine.

✔️ Dosage & Administration of Aristo D3 200000 IU/ml Injection

Injection for Prevention:

Infants receiving Vitamin D enriched milk:

• 1/2 ampoule (0.5ml) i.e. 100,000 I.U. every 6 months.

Nursed infants or infants not receiving Vitamin D enriched milk or young children up to 5 years of age:

• 1 ampoule (1ml) i.e. 200,000 I.U. every 6 months.

Adolescents:

• 1 ampoule (1ml) i.e. 200,000 I.U. every 6 months during winter.

Pregnancy:

• 1/2 ampoule (0.5ml) i.e. 100,000 I.U. from the 6th or 7th month of pregnancy.

Elderly:

• 1/2 ampoule (0.5ml) i.e. 100,000 I.U. every 3 months. For digestive disorders, concomitant treatment with antiepileptics & other particular conditions not described above 1/2 ampoule (0.5ml) i.e. 100,000 I.U. or 1 ampoule (1ml) i.e. 200,000 I.U. every 3 or 6 months.

Injection for Vitamin D Deficiency:

• 1 ampoule (1ml) i.e. 200,000 I.U., which can be repeated 1 to 6 months later, or as directed by the registered physician.

✔️ Overdose Effects of Aristo D3 200000 IU/ml Injection

Hypervitaminosis D may occur.

✔️ Use in Special Populations:

Safety and efficacy in children under 12 years are not established.

✔️ Interaction:

• Increased risk of hypercalcemia with thiazide diuretics, calcium, or phosphate.
• Antiepileptics may increase vitamin D requirements.
• Rifampicin & isoniazid may reduce efficacy.
• Corticosteroids may counteract the vitamin D effect.
• Reduced absorption with cholestyramine, colestipol, mineral oil, orlistat.
• Interaction with digoxin or cardiac glycosides.
• Reduced absorption when taken with ketoconazole.

✔️ Contraindications

Colecalciferol is contraindicated in diseases associated with hypercalcemia, known as hypersensitivity to Colecalciferol, or evidence of vitamin D toxicity.

✔️ Pregnancy & Lactation

Studies suggest safe use during pregnancy up to 4000 IU. Nursing mothers excrete Vitamin D3 in breast milk, but overdosing in infants has not been observed.

✔️ Precautions & Warnings

Monitor plasma calcium concentrations in patients receiving high doses, with renal impairment, or during pregnancy/lactation. Consult healthcare practitioners before use in specific conditions or with certain medications.
