- Prevent bone loss (such as osteoporosis)

- Adults
- Nutritional Deficiency: 5–10 mcg (200–400 IU) daily.
- Osteoporosis (prophylaxis & treatment, >50 years): 800–1000 IU daily with calcium supplements (oral or IM).
- Hypoparathyroidism: 50,000–200,000 IU daily with calcium supplements (oral or IM).
Children
- Nutritional Supplementation:
- 0–12 months: 400 IU daily
- 1–18 years: 600 IU daily
- Vitamin D–Resistant Rickets: 12,000–500,000 IU daily
- Familial Hypophosphatemia: 40,000–80,000 IU daily with phosphate supplements (dose reduced after growth phase).

Hypercalcemia or calcium intoxication
Loss of appetite, headache, dry mouth
Nausea, vomiting, abdominal pain, constipation
✔️ Interaction
Increased hypercalcemia risk: with thiazide diuretics, calcium, phosphate.
Reduced efficacy: with rifampicin, isoniazid, corticosteroids, cholestyramine, colestipol, mineral oil, orlistat, ketoconazole.
Increased requirements: with antiepileptics (carbamazepine, phenytoin, phenobarbital, primidone).
Caution with digoxin/cardiac glycosides: may increase risk of arrhythmias.
✔️ Contraindications:
Hypercalcemia or disorders associated with elevated calcium.
Known hypersensitivity to cholecalciferol or formulation excipients.
Vitamin D toxicity.
✔️ Use with caution in patients with:
- High calcium or phosphorus levels
- Heart disease
- Kidney disorders
- It should be supplemented with adequate calcium and magnesium intake.
- During lactation, vitamin D is secreted into breast milk – use with caution.
