Indication of Tofanib 5mg Tablet

Tofacitinib is indicated for the treatment of adult patients with moderate to severely active rheumatoid arthritis who have not responded or are intolerant to methotrexate. It can be used alone or in combination with methotrexate or other non-biologically modifying anti-rheumatic medications (DMARDs).

Tofacitinib is indicated for the treatment of adult patients with active psoriatic arthritis who have a poor response or intolerance to methotrexate or other disease-modifying antirheumatic drugs (DMARD).

Tofacitinib is approved for the treatment of adult patients with moderate to severe active ulcerative colitis (UC).

Pharmacology

Tofacitinib is a JAK inhibitor that is partially reversible and prevents the body from responding to cytokine signals. Tofacitinib prevents STAT phosphorylation and activation by inhibiting JAK. The JAK-STAT signaling pathway regulates cell transcription in hematopoietic and immune cell functions. Tofacitinib works as an anti-inflammatory agent by inhibiting the JAK-STAT pathway. Dosage & Administration of Tofanib 5mg Tablet
  • Tofacitinib 5mg twice daily or Tofacitinib 11 mg once daily for rheumatoid arthritis. Tofacitinib 5 mg once daily is recommended in patients with moderate to severe renal impairment or moderate Hepatic Impairment.
  • Tofacitinib 5mg twice daily or Tofacitinib 11mg once daily for Psoriatic Arthritis (in combination with nonbiologic DMARDs). Tofacitinib 5mg once daily is recommended in patients with moderate to severe Renal Impairment or moderate hepatic impairment.
  • Tofacitinib 10mg twice daily for at least 8 weeks, followed by 5 or 10mg twice daily. If an adequate therapeutic benefit is not obtained after 16 weeks of 10 mg twice daily, discontinue. To maintain response, use the lowest effective dose.
Interaction
  • Prior to ingesting Tofanib 5mg Tablet If you are taking any of the following medications, let your doctor know:
  • Rifampicin and other antibiotics (used in treating bacterial infections)
    Ketoconazole with fluconazole (used to treat fungal infections)
    Tacrolimus, mercaptopurine, ciclosporin, and azathioprine (medicines that depress the immune system)
  • The use of corticosteroids like prednisone (used in treating pain and inflammation)
Overdosage of Tofanib 5mg Tablet

If you or anyone else unintentionally consumes too much Tofanib 5mg Tablet, call your doctor right away or go to the hospital in the area.

Pregnancy & Lactation

Pregnancy category C. There are no adequate and well-controlled studies of pregnant women. Tofacitinib should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is unknown whether tofacitinib is excreted in human milk.
