✔️ Pharmacology

Insulin lowers blood glucose by helping muscle and fat cells take up glucose and by reducing glucose production by the liver.

Insulet contains two insulin components:

Regular human insulin: Provides the shorter-acting component to help control blood glucose around meals.

Isophane (NPH) insulin: Provides an intermediate-acting component to support blood glucose control between meals and overnight.

The exact action profile varies by formulation and individual response. Because Insulet is premixed insulin, meal timing and consistent administration are important.

✔️ Side Effects:
  • Hypoglycemia (low blood glucose): The most important and common risk.
  • Injection-site reactions, such as redness, swelling, or itching.
  • Lipodystrophy when injection sites are not rotated.
  • Temporary swelling (edema), particularly after starting or changing treatment.
  • Weight gain.
  • Allergic reactions, rarely.
✔️ Directions for Use:

Insulet should not be administered:

During an episode of hypoglycemia.

In patients with known hypersensitivity to human insulin or any ingredient in the formulation.

✔️ Dosage & Administration

The dose must be individualized by a registered physician based on blood glucose levels, meals, physical activity, insulin sensitivity, and other medical conditions.

The required dose may change because of illness, changes in diet or activity, pregnancy, or impaired kidney or liver function. Do not calculate or adjust your dose independently.

How to administer:

  • Insulet is generally injected subcutaneously using the compatible insulin delivery device.
  • Common injection sites include the abdomen, thigh, upper arm, or buttock, depending on the product instructions.
  • Rotate injection sites to reduce the risk of lipodystrophy (changes in fatty tissue under the skin).
  • Follow the specific device instructions for preparing and administering the insulin.
  • The premixed suspension must be resuspended as directed until it appears uniformly white and cloudy. Do not use it if clumps remain or solid particles adhere to the container.
  • Use a new needle for each injection and remove the needle after use according to the device instructions.
  • Follow the prescribed meal schedule. Premixed insulin containing regular insulin generally requires a meal shortly after administration; the exact timing should follow the product label and your clinician's instructions.

Important: Never substitute another insulin brand, strength, or formulation without medical supervision.

✔️ Precautions & Warnings
  • Low blood sugar: Excess insulin, missed or delayed meals, increased physical activity, or alcohol may cause hypoglycemia.
  • Recognize symptoms: Warning signs include sweating, shaking, hunger, dizziness, headache, confusion, and palpitations.
  • Treatment of mild hypoglycemia: If conscious and able to swallowsafely, take fast-acting glucose according to your diabetes care plan and recheck blood glucose after 15 minutes. Repeat if it remains low.
  • Severe hypoglycemia: If a person is unconscious or having a seizure, do not give food or drink by mouth. Use prescribed glucagon if available and someone is trained to administer it, and seek emergency medical assistance.
  • Illness: Fever, infection, surgery, or other acute illness may change insulin requirements. Follow your sick-day plan and seek medical advice.
  • Kidney or liver impairment: Insulin requirements may decrease, increasing the risk of hypoglycemia.
  • Travel: Discuss time-zone changes, meal schedules, and insulin timing with your healthcare professional before travelling.
  • Type 1 diabetes: Do not stop insulin without medical supervision, as this can lead to life-threatening diabetic ketoacidosis.
  • Device and product checks: Confirm the insulin type and strength before every injection.
✔️ Pregnancy & Breastfeeding

Insulin can be used during pregnancy when medically indicated. Maintaining appropriate blood glucose control is important for maternal and fetal health. Insulin requirements may change throughout pregnancy and decrease rapidly after delivery.

Insulin treatment is generally compatible with breastfeeding, although the dose and meal plan may need adjustment. Treatment should be supervised by a healthcare professional.

✔️ Drug Interactions

Other medicines can change insulin requirements.

Medicines that may increase the glucose-lowering effect or hypoglycemia risk include:

  • Other antidiabetic medicines.
  • Monoamine oxidase inhibitors (MAOIs).
  • ACE inhibitors.
  • Salicylates.
  • Some beta-blockers.
  • Alcohol.

Medicines that may increase blood glucose or insulin requirements include:

  • Corticosteroids.
  • Thiazide diuretics.
  • Thyroid hormones.
  • Sympathomimetic medicines.
  • Growth hormone.
  • Danazol.

Beta-blockers may mask some warning signs of hypoglycemia. Octreotide and lanreotide may either increase or decrease insulin requirements. Inform your physician about all medicines and supplements you use.

✔️ Storage:
  • Store unopened insulin according to the manufacturer's instructions, generally refrigerated at 2°C–8°C.
  • Do not freeze insulin or use insulin that has been frozen.
  • Protect from direct sunlight and excessive heat.
  • During use, follow the exact storage temperature and in-use expiry period stated on the Insulet packaging or device instructions.
  • Resuspend the premixed insulin as directed before each injection.
  • Do not use the product if its appearance remains abnormal after proper mixing.
