Glucosamine is a naturally occurring amino sugar that plays an important role in the formation and repair of cartilage. It is a key building block for glycosaminoglycans (GAGs), including hyaluronic acid, keratan sulfate, and chondroitin sulfate, which combine with proteins to form proteoglycans—the structural components of healthy joint cartilage.
As cartilage wears down, particularly in osteoarthritis, glucosamine supplementation helps replenish the body's supply, promoting cartilage repair and supporting the production of connective tissues such as ligaments and tendons. It also stimulates cartilage cells (chondrocytes) to produce more GAGs and proteoglycans.
Following oral administration, approximately 90% of glucosamine sulfate is absorbed through the gastrointestinal tract. It is mainly metabolized by the liver and eliminated through the urine.
Diacerein is an anti-inflammatory medicine used primarily for the treatment of osteoarthritis. It provides pain-relieving and anti-inflammatory effects by inhibiting the production of interleukin-1 (IL-1), a key inflammatory cytokine involved in cartilage breakdown.
By reducing IL-1 activity, Diacerein may slow cartilage degeneration and has demonstrated disease-modifying effects in patients with osteoarthritis, particularly involving the knees and finger joints.
Diacerein has an oral bioavailability of approximately 56%. Taking it with food delays absorption slightly but increases overall absorption by about 25%, making administration with meals preferable. It is extensively metabolized in the liver and excreted mainly in the urine as metabolites.
