Potential side effects include gastrointestinal disturbances, headache, dizziness, changes in liver enzymes, reversible alopecia, rash, fever, blood disorders (e.g., leucopenia, pancytopenia), allergic reactions, convulsions, and meningism in cases of cerebral disease.

How does Estazol 400 work?

The active constituent is converted into its sulfoxide form, leading to the degeneration of cytoplasmic microtubules and tegmental cells. This depletes the parasite's energy and disrupts metabolic processes, ultimately causing the parasite's death.

• Regularly wash your hands with soap and water, especially after using the toilet and while eating.

• Avoid foods that have been kept open in the markets as they can be contaminated.
• Drink boiled and purified water.
• Regular monitoring of blood cell count and liver function is advised during treatment with Albendazole.

Indications:

Estazol is a benzimidazole anthelmintic that is effective against most nematodes and some cestodes. It exhibits vermicidal, ovicidal, and larvicidal activity, primarily by inhibiting tubulin polymerization, leading to the loss of cytoplasmic microtubules in parasites.

Pharmacology:

Albendazole, the active ingredient in Estazol, is a broad-spectrum anthelmintic. It works by blocking glucose uptake in susceptible helminths, depleting their energy reserves and ultimately leading to their death. Albendazole is metabolized in the liver to albendazole sulphoxide, which retains anthelmintic activity. It has a plasma half-life of about 8.5 hours and is excreted primarily in the urine.

Dosage and Administration :

For Adults and Children over 2 years:

• Single-dose treatment: 400 mg (1 tablet or 10 ml suspension) for Enterobius vermicularis, Trichuris trichiura, Ascaris lumbricoides, Ancylostoma duodenale, and Necator americanus.
• For strongyloidiasis or taeniasis: 400 mg daily for 3 consecutive days. A second course may be needed if the patient is not cured after three weeks.

For Children 1-2 years:

• Single-dose treatment: 200 mg (5 ml suspension).

For Children under 1 year:

• Not recommended.

For Hydatid Disease (Echinococcosis):

• For patients over 60 kg: 400 mg twice daily for 28 days.
• For patients under 60 kg: 15 mg/kg daily in two divided doses (maximum total daily dose: 800 mg).
• For cystic echinococcosis, treatment cycles may be repeated after 14 days of no treatment, up to a total of 3 cycles.
• For alveolar echinococcosis, long-term cycles of treatment may be necessary.

For Giardiasis:

• 400 mg once daily for five days.

Interaction

No significant pharmacodynamic or pharmacokinetic interactions have been reported with Estazol.

Contraindications

• Neonates: Not recommended.
• Pregnancy: Should not be used during pregnancy or in women who may be pregnant.
• Children: Dose reduction may be necessary for children weighing less than 10 kg.
• Concurrent Diseases: No dosage adjustment is necessary for renal, hepatic, or cardiac failure patients.

Pregnancy & Lactation

Albendazole falls under the US FDA Pregnancy Category C. It should be avoided during pregnancy and lactation unless the potential benefits to the mother outweigh the risks to the fetus.

Precautions & Warnings

• Regular blood counts and liver function tests are recommended before treatment and during each cycle.
• Ensure pregnancy is excluded before starting treatment.
• Continuous medical supervision is required to treat Echinococcosis, with regular monitoring of serum-transaminase concentrations and blood counts.
