Posaconazole inhibits fungal cell membrane synthesis by blocking ergosterol formation. Specifically, it targets Lanosterol 14-α-demethylase, a cytochrome P450 enzyme responsible for converting lanosterol into ergosterol. This leads to the accumulation of sterol precursors and the depletion of ergosterol, weakening the fungal cell membrane and impairing its function.
