Vitamin D3 (Cholecalciferol) is converted in the liver to 25-hydroxy Vitamin D3 (Calcidiol) and subsequently in the kidneys to the active metabolite, Calcitriol.
Calcitriol acts by binding to Vitamin D receptors (VDRs) throughout the body, increasing intestinal calcium and phosphate absorption necessary for:

- Hypercalcaemia
- Vitamin D toxicity
- Hypersensitivity to Vitamin D3 or formulation ingredients

- Phenytoin and barbiturates
- Glucocorticoids
- Digoxin and cardiac glycosides
- Cholestyramine and paraffin oil
- Antifungal agents and actinomycin
These medicines may reduce Vitamin D absorption or increase calcium-related complications.

- Safe use has been observed at recommended doses during pregnancy
