Glimepiride, a sulfonylurea antidiabetic, works mainly by stimulating insulin secretion from functional pancreatic β-cells.
It enhances β-cell sensitivity to glucose, promoting insulin release in response to physiological glucose levels.
It also improves peripheral insulin sensitivity, increases glucose uptake in tissues, and reduces hepatic glucose production.
A single dose provides 24-hour hypoglycemic control in non-fasting diabetic patients.

Glucorid is used as an adjunct to diet and exercise for reducing blood glucose levels in patients with Type 2 Diabetes Mellitus (NIDDM) whose hyperglycemia is not adequately controlled by lifestyle measures alone.
There are no adequate and well-controlled studies in pregnant women. Glucorid 2 should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus
Avoid consumption of alcohol while taking Glucorid 2, as it may increase the risk of unwanted side effects.
It is not known whether Glucorid 2 is excreted in human milk. Consult your doctor for advice.
Do not drive or operate any heavy tools or machines if your ability is affected by the medicine.
Glucorid 2 is not recommended for use in patients with severe kidney problems. It should be used with caution in patients with other kidney problems.
Glucorid 2 is not recommended for use in patients with severe liver problems. It should be used with caution in patients with other liver problems.
Common: Hypoglycemia, dizziness, nausea, vomiting, abdominal pain
Occasional: Diarrhea, temporary visual disturbance, urticaria, hypotension
Start with 1 mg once daily, taken before breakfast or the first main meal.
If needed, increase gradually every 1–2 weeks based on glucose monitoring:
1 mg → 2 mg → 3 mg → 4 mg → 6 mg daily.
Usual maintenance dose: 1–4 mg daily, depending on glycemic response.
Patients with improved insulin sensitivity may require dose reduction during long-term treatment.
Begin with 1 mg daily even when switching from maximum doses of other agents.
Consider discontinuing the previous drug temporarily to prevent additive hypoglycemic effects.
Swallow tablets with water, just before or with meals.
Avoid skipping meals after taking the dose.
Insulin, metformin, ACE inhibitors, anabolic steroids, azapropazone, chloramphenicol, ciprofibrate, fluconazole, fluoxetine, coumarins, disopyramide, fenfluramine, MAO inhibitors, miconazole, quinolones, salicylates, sulfonamides, and tetracyclines.
Corticosteroids, diuretics, phenytoin, thyroid hormones, calcium channel blockers, barbiturates, nicotinic acid (high dose), estrogen, rifampicin, and sympathomimetics.
Beta-blockers, clonidine, and reserpine may either enhance or reduce hypoglycemic activity and can mask early symptoms of low blood sugar.
Alcohol intake may unpredictably alter the effect of Glucorid.
Contraindicated in pregnancy: Switch to insulin for glycemic control.
Breastfeeding: Avoid use — Glimepiride may pass into breast milk and harm the infant.
Discontinue breastfeeding or use alternative therapy as advised by a physician.
