Vonoprazan reversibly inhibits the H⁺/K⁺-ATPase in gastric parietal cells through a potassium-competitive mechanism. Unlike conventional proton pump inhibitors, it does not require activation by gastric acid. Its strong and prolonged action at the acid-secreting site of gastric parietal cells produces potent suppression of gastric acid secretion and helps protect against acid-related damage in the upper gastrointestinal tract.

✅ Description:
Gastric and Duodenal Ulcers
- Gastric ulcer: 20 mg once daily for 8 weeks.
- Duodenal ulcer: 20 mg once daily for 6 weeks.
Reflux Esophagitis
- Treatment: 20 mg once daily for 4 weeks. If the response is inadequate, treatment may be extended, but not beyond 8 weeks.
- Maintenance therapy for recurrent reflux esophagitis: 10 mg once daily.
- If 10 mg is insufficient, the dose may be increased to 20 mg once daily.
Prevention of Ulcer Recurrence
For patients receiving:
- Low-dose aspirin: 10 mg once daily.
- NSAIDs: 10 mg once daily.
Helicobacter pylori Eradication
Initial three-drug regimen: Take all three medicines twice daily for 7 days:
- Vonoprazan 20 mg
- Amoxicillin hydrate 750 mg
- Clarithromycin 200 mg
The clarithromycin dose may be increased when clinically appropriate, but should not exceed 400 mg twice daily.
Alternative regimen after unsuccessful PPI-based eradication therapy:
- Vonoprazan 20 mg
- Amoxicillin hydrate 750 mg
- Metronidazole 250 mg
Take all three medicines twice daily for 7 days.
Take the medicine only as directed by your doctor.

Vonoprazan can be taken with or without food and does not need to be taken at a specific time in relation to meals.
- Elderly patients: Use with caution because liver and kidney function may be reduced.
- Children and adolescents below 18 years: Safety and efficacy have not been established; use is not recommended.
✔️ Vonoprazan should not be used in patients with:
- Hypersensitivity to vonoprazan or any component of the formulation.
- Concomitant use of atazanavir sulfate, nelfinavir, or rilpivirine hydrochloride.

- Treatment should be limited to the minimum duration necessary according to the patient's condition.

- Maintenance therapy for reflux esophagitis should generally be reserved for patients with recurrent disease.
- If healing remains stable for a prolonged period and the risk of recurrence is low, reducing the dose from 20 mg to 10 mg or discontinuing treatment may be considered.
- Use should follow the advice of a registered physician.
