Adalimumab is a fully human monoclonal IgG1 antibody targeting TNF-alpha. It binds with high specificity to TNF-alpha, preventing its interaction with cell surface TNF receptors (p55 and p75). In vitro, it can also mediate lysis of cells presenting surface TNF in the presence of complement. TNF-alpha is an inflammatory cytokine playing a key role in immune system regulation. Elevated TNF-alpha levels are associated with inflammatory conditions such as RA, JIA, PsA, AS, and psoriasis. In these conditions, excessive TNF contributes to inflammation and joint/tissue damage. Adalimumab’s mechanism helps reduce inflammation, joint erosion, and skin plaque thickness.

Pediatric Crohn’s Disease:
17 to <40 kg:
Day 1: 80 mg (2 injections)
Day 15: 40 mg
Day 29 onwards: 20 mg every other week
≥40 kg:
Day 1: 160 mg (4 injections or 2 per day over 2 days)
Day 15: 80 mg (2 injections)
Day 29 onwards: 40 mg every other week
Abatacept / Anakinra: Increases the risk of severe infections—concurrent use should be avoided.
Live vaccines: Contraindicated while on Advixa due to the risk of infection.
Known allergy or hypersensitivity to adalimumab or its formulation components.
Pregnancy: Classified as Category B. Animal studies haven’t shown harm, but there are no adequate studies in humans. As an IgG1 antibody, adalimumab crosses the placenta, especially in the third trimester.
Lactation: Small amounts may pass into breast milk, but absorption by the infant is unlikely. Use caution in nursing mothers
