Rupatadine acts as a long-acting, non-sedative antagonist of histamine H1-receptors. Additionally, it antagonizes the platelet-activating factor (PAF). Both histamine and PAF contribute to bronchoconstriction, leading to increased vascular permeability and acting as mediators in the inflammatory process. Rupatadine's dual mode of action results in a better therapeutic effect compared to isolated antihistamines. Furthermore, Rupatadine possesses other anti-allergic properties, including inhibiting the degranulation of mast cells induced by both immunological and non-immunological stimuli. It also inhibits the release of cytokines, particularly tumor necrosis factor alpha (TNF α), in human mastocytes and monocytes.
For adults and adolescents over 12 years old, the recommended dose of Rupatadine is 10 mg once daily. It can be taken with or without food.
