Pharmacology
  • Tacrolimus is derived from the fungus Streptomyces tsukubaensis.
  • Tacrolimus acts as a macroiide immunomodulator, primarily by inhibiting T-lymphocyte activation.
  • Initially, Tacrolimus binds to an intracellular protein called FKBP-12.
  • The Tacrolimus-FKBP-12 complex, along with calcium, calmodulin, and calcineurin, forms a complex.
  • This complex inhibits the phosphatase activity of calcineurin, a critical enzyme in T-cell activation.
  • By inhibiting calcineurin activity, Tacrolimus disrupts the entire inflammatory cascade, resulting in a decrease in inflammation.
