✔️ Dosage:

Defrol can be taken with or without food. It may be administered orally or intramuscularly (IM) as prescribed by the healthcare provider.

Adult Dosage

1. Nutritional Deficiency (Vitamin D3):
   • 5-10 mcg (200-400 IU) per day orally.
2. Osteoporosis Prophylaxis and Treatment (>50 years):
   • 800-1000 IU (20-25 mcg) daily with calcium supplements.
   • May also be given as an IM injection.
3. Hypoparathyroidism:
   • 50,000-200,000 IU (0.625-5 mg) orally once daily with calcium supplements, or via IM injection.

Child Dosage

1. Nutritional Supplementation:
   • 0-12 months: 400 IU (10 mcg) once daily.
   • 1-18 years: 600 IU (15 mcg) once daily.
2. Vitamin D-Resistant Rickets:
   • 12,000-500,000 IU (0.3-12.5 mg) orally once daily.
3. Familial Hypophosphatemia:
   • 40,000-80,000 IU (1-2 mg) once daily with phosphate supplements.
   • May be reduced after the growth phase is complete.

✔️ Generally well tolerated, but some side effects may include:

• Hypercalcaemia (excessive calcium) or calcium intoxication.
• Acute symptoms of hypercalcemia: Anorexia, headache, dry mouth, nausea, vomiting, abdominal pain, or constipation.

✔️ Colecalciferol should not be used in:

• Patients with hypercalcemia (high blood calcium levels).
• Those with known hypersensitivity to Colecalciferol or related medications.
• Vitamin D toxicity.

✔️ Mode of Action:

Colecalciferol is a fat-soluble sterol essential for calcium and phosphate homeostasis, as well as bone mineralization. It enhances calcium absorption from the gastrointestinal tract and stimulates bone formation, promoting overall bone health.

✔️ Interactions

• Increased risk of hypercalcaemia when used with thiazide diuretics, calcium, or phosphate.
• Antiepileptic medications (e.g., carbamazepine, phenobarbitone, phenytoin, primidone) may require higher doses of vitamin D.
• Rifampicin and isoniazid may reduce vitamin D efficacy.
• Corticosteroids may counteract Vitamin D effects.
• Digoxin or any cardiac glycosides may have interactions.
• Reduced absorption when taken with cholestyramine, colestipol, mineral oil, or orlistat

✔️ Use with caution in individuals with:

• High blood calcium or phosphorus levels.
• Heart problems.
• Kidney disease.
• Ensure adequate intake of calcium and magnesium alongside Vitamin D.
• Lactation: Vitamin D is distributed into breast milk; use cautiously during breastfeeding.
