How to use Amilin 25

Take this medicine in the dose and duration as advised by your doctor. Swallow it as a whole. Do not chew, crush or break it. Amilin 25 may be taken with or without food, but it is better to take it at a fixed time.

How does Amilin 25 work?

Indications of Amilin 25

Amilin 25 is used to treat mental/emotional issues including depression. It may help you boost your mood and sense of well-being, decrease worry and stress, sleep better, and have more energy. This drug belongs to the tricyclic antidepressant family of pharmaceuticals. It works by changing the balance of certain natural chemicals in the brain (neurotransmitters like serotonin).

Pharmacology

The mechanism of action of this drug is not fully understood. Amitriptyline is thought to inhibit the membrane pump mechanism, which allows transmitter amines such as norepinephrine and serotonin to re-enter the brain, thereby increasing their concentration in synaptic clefts. These amines are essential for mood regulation. One of the oldest theories about depression is that depression is caused by a lack of serotonin (5-HT) and/or norepinephrine (NE) neurotransmission in the brain.

Dosage & Administration of Amilin 25

Depression: 

• 75 mg (Elderly and Adolescents 30-75 mg) daily in divided doses or as a single dose at bedtime, gradually increased to 150-200 mg as needed;
• Children under the age of 16 are not recommended for depression.

Nocturnal Enuresis: 

• 10-20 mg for children aged 7 to 10, 25-50 mg

For Children aged 11 to 16:

• With a maximum dose of 100 mg. Treatment period (including gradual withdrawal) 3 months-full physical examination before the next course

Migraine Prophylaxis: 100 mg daily.

• 10-25 mg three times per day for Tension Headaches.

Interaction

Alcohol and opioid analgesics have a stronger sedative impact when combined with TCA. TCA raises the risk of ventricular arrhythmias when used with Moxifloxacin or Terfenadine. Amitriptyline metabolism is inhibited by Disulfiram and Cimetidine. TCA increases the risk of postural hypotension when used with diuretics.

Contraindications

Myocardial infarction, arrhythmias, especially heart block of any degree, mania, and severe liver illness are all contraindications to taking amitriptyline. Sedation might impair one's ability to drive or operate machines at first.

Pregnancy & Lactation

• Pregnancy Classification C. It has been proven that amitriptyline crosses the placenta. During pregnancy, amitriptyline should only be taken if the possible benefit to the mother outweighs the danger to the fetus.
• Amitriptyline is passed through the breast milk. Because Amitriptyline has the potential to produce significant adverse effects in nursing infants, a choice should be taken whether to stop breastfeeding or stop taking the medicine, taking into consideration the drug's value to the mother.

Precautions & Warnings

• In individuals with a history of epilepsy, glaucoma, urinary retention, heart illness, diabetes, pregnancy, hepatic impairment, thyroid disease, elevated intra-ocular pressure, or psychoses, it should be taken with caution (may aggravate mania).
