• Aproxi 80 is used alongside other medications to prevent nausea and vomiting following chemotherapy. Even if you do not feel like eating or drinking, it is important to sip water regularly to avoid dehydration.
• If you are taking oral contraceptives while using this medication, its effectiveness may be reduced if you experience prolonged vomiting lasting more than 24 hours. In such cases, consult your doctor for additional contraceptive measures.
• Aproxi 80 is effective in preventing nausea and vomiting caused by certain chemotherapy drugs but does not treat existing symptoms. It should not be used continuously for an extended period.
• It is advised to take Aproxi 80 one hour before chemotherapy on the first day and in the morning on days two and three.
• Additionally, Aproxi 80 may impact sperm health and pose risks during pregnancy or breastfeeding.

✔️ Aprepitant Dosage Guidelines

Adult Dose

Prevention of Postoperative Nausea and Vomiting (PONV)

• 40 mg orally within 3 hours before induction of anesthesia.

Prevention of Chemotherapy-Induced Nausea and Vomiting (CINV)

• Day 1: 125 mg orally, 1 hour before chemotherapy
• Days 2 & 3: 80 mg orally in the morning
• Must be administered with a corticosteroid (e.g., dexamethasone) and a 5-HT₃ antagonist (e.g., ondansetron).

Use with caution in patients with moderate to severe liver impairment.

Child Dose

• Children under 12 years or weighing less than 30 kg: Safety and efficacy not established.

• Increased Aprepitant Levels: Co-administration with CYP3A4 inhibitors (e.g., clarithromycin, troleandomycin, diltiazem, verapamil, itraconazole, ketoconazole, nefazodone, ritonavir, nelfinavir) can increase plasma levels of Aprepitant.
• Decreased Aprepitant Efficacy: Co-administration with CYP3A4 inducers (e.g., carbamazepine, phenytoin, rifampicin, primidone, phenobarbital) may reduce the effectiveness of Aprepitant.
• Effect on Other Drugs:
  • May reduce the levels of CYP2C9 substrates such as phenytoin, tolbutamide, and warfarin.
  • May increase plasma concentrations of dexamethasone and methylprednisolone.
  • May increase levels of benzodiazepines like midazolam, diazepam, and alprazolam.
  • May increase serum concentrations of colchicine.
  • May increase toxicity of chemotherapy drugs metabolized by CYP3A4 (e.g., docetaxel, etoposide, ifosfamide, imatinib, paclitaxel, vincristine, vinblastine).
  • May increase the levels of ergot derivatives, leading to ergotism.
  • May reduce the effectiveness of oral contraceptives, necessitating additional contraceptive measures for at least one month after the last dose.

Aprepitant may elevate the serum levels of astemizole, cisapride, pimozide, and terfenadine, which could lead to life-threatening torsade de pointes arrhythmias.

✔️ Drug Warnings: Aproxi 80

Inform your doctor if you are allergic to Aprifast 125/80 mg Capsule Kit or any other medications. This medicine is not recommended for children under 12 years old. If you are pregnant or breastfeeding, consult your doctor before use. Avoid driving or operating machinery if you experience dizziness or drowsiness after taking Aprifast 125/80 mg Capsule Kit. If you are using any other prescription or non-prescription drugs, inform your doctor before starting this medication.

Pregnancy: There is insufficient data on the use of this drug in pregnant women to determine any associated risks. Animal studies have not shown adverse developmental effects at exposure levels similar to those in humans. However, due to the alcohol content in the intravenous (IV) form of Aprepitant, its use should be avoided during pregnancy.

Lactation: It is unclear whether Aprepitant is excreted in human milk. A decision should be made to either discontinue breastfeeding or discontinue the medication, depending on its importance to the mother.
