Ketoprofen is a non-steroidal anti-inflammatory drug (NSAID) with analgesic and antipyretic effects. It works by inhibiting prostaglandin synthesis and stabilizing lysosomal membranes. Additionally, it inhibits leukotriene synthesis at high concentrations and exhibits antibradykinin activity. Ketoprofen provides pain relief by blocking prostaglandin synthesis both centrally and peripherally. It may also suppress prostaglandin synthesis in the central nervous system, particularly in the hypothalamus, to exert its antipyretic effect.
Ketoprofen is absorbed rapidly from the gastrointestinal tract and is highly protein-bound (approximately 99%). Its elimination half-life is 1.1 to 4 hours and is primarily metabolized in the liver through conjugation with glucuronic acid. Most of the drug is excreted via urine (50–90%) and feces (1–8%) within 1–5 days. After intramuscular injection, the peak concentration is reached within 0.5 to 0.75 hours.
