✅ Description:
Pazopanib is a multi-tyrosine kinase inhibitor targeting various receptors, including:
- VEGFR-1, VEGFR-2, VEGFR-3 (vascular endothelial growth factor receptors)
- PDGFR-α and PDGFR-β (platelet-derived growth factor receptors)
- FGFR-1 and FGFR-3 (fibroblast growth factor receptors)
- Kit, Itk, Lck, and c-Fms (cytokine and protein tyrosine kinases).
Pazopanib works by inhibiting the autophosphorylation of several growth factor receptors, thereby exerting its anti-cancer effects.
Absorption
Pazopanib is absorbed orally, reaching peak concentration in 2 to 4 hours. When administered at 800 mg daily, the geometric mean AUC (Area Under the Curve) is 1,037 mcg/mL and C max is 58.1 mcg/mL.
Distribution
Pazopanib binds to plasma proteins in humans with an in vivo binding of greater than 99%, unaffected by concentration over the range of 10 to 100 mcg/mL.
Metabolism
Pazopanib is metabolized primarily by CYP3A4, with some contributions from CYP1A2 and CYP2C8.
Elimination
The drug has a half-life of 30.9 hours after an 800 mg dose. The majority is eliminated via feces, with less than 4% excreted renally.

✔️ Dosage & Administration
- Recommended starting dose: 800 mg orally once daily, on an empty stomach (at least 1 hour before or 2 hours after a meal).
- Maximum dose: 800 mg daily.
- Tablets should not be crushed due to the risk of increased absorption.
- If a dose is missed and less than 12 hours remain until the next dose, skip the missed dose. Do not take a double dose.
- Females: Should use effective contraception during treatment and for at least 2 weeks after the last dose of Pazocent.
- Males: Should use condoms during treatment and for at least 2 weeks after the last dose to avoid exposure to pregnant partners.
- Pediatric Use: Safety and efficacy in pediatric patients have not been established.
✔️ Interactions
- CYP3A4 Inhibitors (e.g., Ketoconazole, Ritonavir, Clarithromycin) may increase Pazocent concentrations. If co-administered, reduce the dose to 400 mg.
- CYP3A4 Inducers (e.g., Rifampin) may decrease Pazocent concentrations.
- P-glycoprotein (P-gp) and BCRP inhibitors should be avoided, as they may increase Pazocent exposure.
- Pazocent weakly inhibits CYP3A4, CYP2C8, and CYP2D6, which may affect drugs metabolized by these enzymes.

Pazocent is contraindicated in patients who are hypersensitive to Pazopanib or any of its components.
✔️ Pregnancy & Lactation
- Pregnancy: Pazopanib can harm the fetus. It is advised that women of reproductive potential use effective contraception during treatment and for at least 2 weeks after the final dose.
- Lactation: There is no information on whether Pazopanib is excreted in human milk. Due to potentially serious side effects in breastfed infants, breastfeeding should be avoided during treatment and for 2 weeks after the last dose.
✔️ Precautions & Warnings
- Hepatic Toxicity: Hepatotoxicity, including severe and fatal liver damage, has been observed, especially in patients over 65.
- QT Prolongation: Pazocent may prolong the QT interval, particularly in patients with a history of QT prolongation. Monitoring of ECG and electrolytes is recommended.
