Defera is used to manage chronic iron overload in patients with thalassemia major when deferoxamine is inadequate or not tolerated. Treatment should be supervised by a physician experienced in managing iron overload conditions.
Deferiprone is an oral iron chelator with high selectivity for ferric iron (Fe³⁺). It forms stable 3:1 (Deferiprone:Iron) complexes that are efficiently excreted, reducing excess body iron. It shows lower affinity for other metal ions like copper, zinc, and aluminum.
Gastrointestinal discomfort: nausea, vomiting, diarrhea, abdominal pain
Reddish-brown urine discoloration (harmless)
Other possible effects:
Adults & children >6 years: 25 mg/kg three times daily (total max: 100 mg/kg/day)
Dosing must be individualized and supervised by a physician
Take orally, with or without food
Avoid doses exceeding the recommended maximum
Caution with Vitamin C: May increase risk of adverse effects (as seen with deferoxamine)
Avoid use with aluminum-containing antacids
Known hypersensitivity to deferiprone or any component of the formulation
Not recommended during pregnancy or breastfeeding due to potential fetal or infant risk.
Monitor complete blood count (especially neutrophils) weekly
Use with caution in patients with hepatic or renal impairment
Store below 30°C, away from moisture and light.
Keep out of reach of children.
