✔️ Pharmacology

Clomipramine is a potent inhibitor of serotonin re-uptake in the brain. It has significant antagonism at cholinergic and α1-receptors and weak antagonism at dopamine receptors, providing antidepressant, sedative, and anticholinergic effects.

✔️ Dosage & Administration:

The initial dose is 25 mg at bedtime for about a week. Gradually increase the dose, ensuring it does not exceed 150 mg per day for adults and 75 mg per day for elderly patients. The dosage can be divided to minimize gastrointestinal side effects. 

Not recommended for children. 

Take Clofranil 25mg Tablet with food, as advised by your doctor. Swallow the tablet whole with water. If you miss a dose, take it as soon as you remember. If it’s almost time for your next dose, skip the missed dose. Do not double the dose.

✔️ Interaction

• Barbiturates increase the metabolism of tricyclic antidepressants.
• Cimetidine, guanethidine, haloperidol, and phenothiazines block tricyclic metabolism.
• Enhanced CNS effects with alcohol.
• MAOIs require a gap of at least 3 weeks before switching to Clomipramine.
• Risk of hypertension and arrhythmias if combined with adrenaline and noradrenaline.

• Hypersensitivity to Clomipramine Hydrochloride or other tricyclic antidepressants in the dibenzazepine group
• Recent myocardial infarction recovery phase
• Concomitant use of MAO inhibitors
• Heart block or other arrhythmias
• Mania

✔️ Pregnancy & Lactation

No adequate well-controlled studies of safety for pregnancy. Limited data suggest that clomipramine is likely to be excreted in human breast milk. Although it has known risk of damage to fetus.
