How does Chlorambucil work?

Chlorambucil interferes with DNA replication and damages DNA in cells. This DNA damage induces cell cycle arrest and cellular apoptosis, ultimately leading to the death of cancer cells.

Side Effects:

Common side effects include bone marrow suppression, nausea, vomiting, diarrhea, and oral ulceration. Rare side effects may include irreversible bone marrow failure, allergic reactions, Stevens-Johnson syndrome, seizures, and hepatotoxicity.

Administration :

Leukeran Tablet should be taken on an empty stomach, ensuring adequate hydration. It should be swallowed whole without chewing or crushing.

Dosage

The dosage varies based on the condition being treated:

• Hodgkin's disease: 200 mcg/kg/day for 4-8 weeks.
• Non-Hodgkin's lymphoma: 100-200 mcg/kg/day for 4-8 weeks.
• Chronic lymphocytic leukemia: Initially, 150 mcg/kg/day until the total leukocyte count falls to 10,000 microliters, then resumed at 100 mcg/kg/day.
• Waldenstrom's macroglobulinemia: Starting dose of 6-12 mg daily until leucopenia occurs, followed by 2-8 mg daily.

Pregnancy & Lactation

Leukeran Tablet is contraindicated during the first trimester of pregnancy and lactation due to the potential risks to the fetus or infant. However, in certain life-threatening situations, the benefits may outweigh the risks.

Precautions & Warnings

Close monitoring of full blood count (FBC) is recommended, and bone marrow suppression is reversible if treatment is stopped early. Caution is advised in patients with renal or hepatic impairment, seizure disorders, and those undergoing high pulse dosing regimens. Adequate contraceptive precautions should be ensured during treatment.
