How does Paloxi 0.5mg Tablet work?

Palonosetron, the active ingredient in Paloxi 0.5mg Tablet, belongs to a group of drugs called serotonin antagonists. These drugs work by blocking the action of serotonin, which is a chemical that transmits messages between nerve cells in the brain and throughout the body. By blocking serotonin, Paloxi 0.5mg Tablet helps to control nausea and vomiting caused by chemotherapy in adults.

Indication of Paloxi 0.5mg Tablet

Palonosetron is used to treat-

• For postoperative nausea and vomiting following surgery

Pharmacology

Palonosetron is a medication that blocks the 5-HT3 receptor, which is responsible for causing nausea and vomiting when stimulated by serotonin released from cells in the small intestine due to chemotherapy. This drug has a strong affinity for the 5-HT3 receptor and does not affect other receptors.

Dosage & Administration of Paloxi 0.5mg Tablet

• This medication is used for the prevention of nausea and vomiting caused by chemotherapy or after surgery. In adults, it can be given as a single 0.25 mg IV dose over 30 seconds, 30 minutes before chemotherapy, and should not be repeated within 7 days. It can also be taken orally as a 0.5 mg tablet/capsule approximately 1 hour before chemotherapy, followed by a 0.5 mg tablet once daily.
• For children between 1 month and 17 years of age, the recommended dose is 20 mcg/kg IV infused over 15 minutes, starting 30 minutes before chemotherapy, not to exceed 1.5 mg/dose. It is not recommended for children under 1 month of age.
• This medication does not require dose adjustment for patients with hepatic or renal impairment.

Paloxi 0.5mg Tablet: Place the tablet in your mouth and do not swallow it. Allow it to melt completely. Do not handle the tablet with wet hands.

Interaction

Paloxi 0.5mg Tablet may interact with several drugs including antidepressants, heart-related drugs or high blood pressure medications, antibiotics, antiemetics, and antipsychotic drugs. Patients with blocked bowel, repeated constipation, heart problems, or imbalance of potassium and magnesium in the blood should inform their doctor before taking Paloxi 0.5mg Tablet. It is important to let your doctor know about any other medications or supplements you are taking to prevent any potential drug interactions.

Contraindications

Palonosetron is not recommended for patients who have a history of hypersensitivity to the drug or any of its ingredients.

Pregnancy & Lactation

• Palonosetron is not recommended for use in pregnancy unless clearly necessary as it is not known whether it may cause harm to the unborn baby. Contact your doctor before taking it.
• It is not known whether Palonosetron passes through breast milk, hence consult your doctor before taking this medicine

Precautions & Warnings

• When hypercalcemia occurs, the drug must be stopped immediately.
• Paloxi 0.5mg Tablet should not be used in patients who have intestinal obstruction or ileus as it may worsen the condition. Rapid injection of the medicine may also cause temporary visual changes like blurred vision.
• Patients who have or may develop prolonged QT intervals should use Paloxi 0.5mg Tablet with caution as it may worsen the condition.
• The safety and efficacy of Paloxi 0.5mg Tablet have not been established in children under the age of 18.
• Increased fluid intake should also be recommended for patients who have a history of kidney stone formation.
• It works quickly and begins working within 30 minutes.
• Take another dose if you vomit within one hour of taking one.
• Avoid large meals and instead, opt for small nourishing snacks throughout the day.
• Drink water on a regular basis to avoid dehydration.
