Olanzapine is an antipsychotic medication that interacts with multiple receptors, including serotonin (5HT2A/2C, 5HT3, 5HT6), dopamine (D1-D5), muscarinic (M1-M5), adrenergic (α1), and histamine (H1). While its exact mechanism in treating schizophrenia is not fully understood, it is believed to work by blocking dopamine and serotonin (5HT2) receptors. Olanzapine is efficiently absorbed after oral intake, reaching peak levels in 5-8 hours, and food intake does not affect its absorption. Studies have found that Olanzapine does not cause genetic mutations, chromosomal damage, or cancer.

Oleanz is prescribed for both acute and long-term treatment of schizophrenia and related psychotic disorders, including symptoms such as delusions, hallucinations, disorganized thinking, hostility, and paranoia, as well as negative symptoms like emotional withdrawal, reduced speech, and lack of social engagement. It is also used to manage acute manic or mixed episodes in bipolar disorder, with or without psychotic features or rapid cycling.

✔️ Side Effects
Very common side effects include drowsiness and weight gain. Other possible effects include increased appetite, elevated glucose and triglyceride levels, dizziness, restlessness (akathisia), Parkinsonism, involuntary muscle movements (dyskinesia), low blood pressure (orthostatic hypotension), constipation, dry mouth, fatigue, swelling, and sensitivity to sunlight.

- Schizophrenia & Bipolar Disorder (Combination Therapy for Mania): Initial dose of 5-10 mg once daily, adjusted as needed within 5-20 mg/day. Doses exceeding 10 mg should be used only after reassessment. Maximum dose: 20 mg/day.
- Mania (Monotherapy): Starting dose of 15 mg once daily, with possible adjustments between 5-20 mg/day. Doses above 15 mg should only be considered after reassessment. Maximum dose: 20 mg/day.
Use this medication only as directed by a registered physician. - against operating heavy machinery or driving due to potential drowsiness.
- Children: Not recommended for individuals under 18 years.
- Elderly (65+): Starting dose of 5 mg/day.
- Patients with liver or kidney impairment: Initial dose of 5 mg/day.

- May counteract the effects of levodopa and dopamine agonists.
- Medications that induce CYP1A2 or glucuronyl transferase (e.g., Omeprazole, Rifampicin) may accelerate Olanzapine clearance.
- CYP1A2 inhibitors may slow down Olanzapine elimination.
- Carbamazepine may increase Olanzapine clearance.
- Activated charcoal reduces Olanzapine absorption by 50-60%.
- Caution is advised when taken with alcohol or other central nervous system depressants.

Not recommended for patients with known hypersensitivity to Olanzapine or those at risk of narrow-angle glaucoma.
