✔️ Common Side Effect

Sporium can cause a range of side effects. Common ones include:

• Headache
• Nausea or vomiting
• Tremors
• Diarrhea
• Loss of appetite
• High blood pressure
• Increased hair growth (hirsutism)

Serious and Rare Side Effects

• Infections due to immune suppression
• Kidney toxicity
• Liver problems
• High potassium levels
• Seizures
• Allergic reactions, including anaphylaxis (rare)
• Abnormal bleeding or bruising
• Gum overgrowth

✔️ Dosage Guidelines

• Dosage varies depending on the condition being treated:
• Organ Transplantation (Prophylaxis): Start with 10–15 mg/kg/day, 4–12 hours before surgery; maintenance is usually 2–6 mg/kg/day.
• Rheumatoid Arthritis: Typically 2.5 mg/kg/day in divided doses; may be increased up to 4 mg/kg/day.
• Psoriasis: Start with 2.5 mg/kg/day, up to a maximum of 5 mg/kg/day if needed.
• Nephrotic Syndrome: Adults—5 mg/kg/day; Children—6 mg/kg/day in divided doses.
• Severe Atopic Dermatitis: Start with 2.5 mg/kg/day; discontinue if no improvement by 6 weeks.
• Bone Marrow Transplantation (IV): 3–5 mg/kg/day starting the day before transplant, then switch to oral maintenance at 12.5 mg/kg/day.

✔️ Drug Interactions

Sporium interacts with many medications:

• Increases blood levels: Diltiazem, erythromycin, ketoconazole, verapamil, oral contraceptives, and methylprednisolone.
• Reduces blood levels: Carbamazepine, phenytoin, phenobarbital, rifampin, isoniazid.
• Risk of toxicity: When used with nephrotoxic drugs like NSAIDs, aminoglycosides, or amphotericin B.
• Potentially fatal interactions: Co-administration with high-dose steroids may lead to seizures.

✔️ Contraindications

Sporium should not be used in:

• Individuals allergic to cyclosporine or any excipient in the formulation.
• Patients with uncontrolled hypertension, active infections, or malignancies (particularly skin cancers).
• Pregnant or breastfeeding women, unless absolutely necessary.

✔️ Special Precautions

• Cancer Risk: Long-term use increases the risk of lymphomas and skin cancers. Avoid excessive sun exposure.
• Kidney Function: May cause renal impairment. Monitor serum creatinine and urea regularly.
• Blood Pressure: Monitor and manage any rise in blood pressure.
• Infections: Higher risk due to immune suppression—monitor for signs and symptoms.
• Electrolyte Imbalance: Risk of hyperkalemia and hypomagnesemia—monitor electrolytes.
• Vaccination: Live vaccines should be avoided; immune response may be diminished.
• Elderly: Require closer monitoring, especially of kidney function.
• Pregnancy Category C: Risk to fetus cannot be ruled out; only use if clearly needed.
• Breastfeeding: Not recommended as cyclosporine is excreted in breast milk.
