Indications of Dinogest 2

The drug Dinogest 2 is used to treat endometriosis-related pelvic pain. It contains progestin, which functions by reducing the endometrium's hormone-producing ability. Progestin is identical to the female hormone.

Pharmacology

Dienogest has a very powerful progestogenic activity in the endometrium but is just a mild agonist at the progesterone receptor (PR) with a similar affinity to progesterone, leading to endometrial atrophy with continued use. It has antiproliferative, immunologic, and antiangiogenic effects on endometrial tissue. By reducing endogenous oestradiol synthesis, dienogest inhibits the trophic effects of oestradiol in both eutopic and ectopic endometrium. Dienogest treatment continuously produces a hyper-progestogenic and moderately hypoestrogenic endocrine milieu that hastens the decidualization of endometrial tissue.

Dosage & Administration

Starting to take tablets is possible at any point in the menstrual cycle. Dienogest should be taken at a dose of 2 mg daily, without a break, preferably at the same time each day with any necessary liquids. It is necessary to take the tablet consistently regardless of vaginal bleeding. When a pack is done, the subsequent one should immediately begin.

If the woman forgets to take a tablet, she should take 2 mg as soon as she remembers, and she should resume taking the tablet at her regular time the next day. Likewise, a 2 mg tablet should be substituted for any that were not absorbed due to nausea or diarrhea.

Interaction of Dinogest 2

Like other progestogens, dienogest is mostly metabolized by the cytochrome P450 3A4 system. Therefore, progestogen drug metabolism may be impacted by CYP3A4 inducers or inhibitors. Verapamil, diltiazem, erythromycin, clarithromycin, and roxithromycin are examples of macrolides. Protease inhibitors include ritonavir, saquinavir, indinavir, and nelfinavir. Antidepressants include nefazodone and fluvoxamine.

Contraindications

hypersensitivity to any of the excipients in dienogest or to dienogest itself. In the event of any of the following conditions, dienogest should not be used: the presence or history of severe hepatic disease as long as liver function values have not returned to normal; the presence or history of liver tumors (benign or malignant); active venous thromboembolic disorder; arterial and cardiovascular disease (such as myocardial infarction, cerebrovascular accident, ischemic heart disease); diabetes mellitus with vascular involvement; a hormone-related sexual disorder that is either known or suspected.

Side Effect of Dinogest 2

Unwanted effects are more prevalent in the first several months after starting Dienogest therapy, although they fade over time. Dienogest users have reported the following negative side effects. Headache (9 percent), breast pain (5.4 percent), low mood (5.1 percent), and acne were the most often reported negative effects during therapy that were thought to be at least partly linked to Dienogest (5.1 percent ).

Headaches and migraines are examples of nervous system disorders.

Uncommon cardiac disorders include palpitations and unspecified circulatory system disease.

Hypotension is a rare vascular disorder.

Nausea, stomach discomfort, and flatulence are common gastrointestinal disorders.

Disorders of metabolism and nutrition: gaining weight (3.6 percent )

Depressed mood, anger, anxiety, and changed mood are all psychiatric disorders.

Pregnancy & Lactation

Dienogest's usage in pregnant women has been studied just a little. Pregnancy, embryonic/fetal development, birth, and development after delivery are all unaffected in humans, according to animal studies and data from women exposed to Dienogest during pregnancy. Dienogest should not be given to pregnant women since endometriosis does not need to be treated during pregnancy.

Dienogest should not be used during pregnancy or breastfeeding. Dienogest is excreted in breast milk, according to physiochemical characteristics and animal studies.
