Semaglo is a GLP-1 receptor agonist that reduces blood glucose levels by stimulating insulin secretion and lowering glucagon secretion when blood glucose levels are high. It also delays gastric emptying and reduces appetite, leading to weight loss.

✔️ Quick Suggestions:

• Take Semaglutide as advised by your physician. It should be taken on an empty stomach. After taking Semaglutide wait at least 30 minutes before having your first meal or drink of the day or taking other oral medicines. Do not stop using this medicine without telling your doctor. Otherwise, your blood sugar levels may raise
• Stay hydrated by drinking plenty of water throughout the day
• Attend regular health checkups while taking this medicine
• Inform your healthcare provider about all the medications, supplements, and herbal products you are using, as Semaglutide can interact with certain drugs
• Semaglutide is not recommended for use in pregnant or breastfeeding women

✔️ Indication of Semaglo Subcutaneous Injection

Semaglo Subcutaneous Injection is indicated as an adjunct to diet and exercise to improve glycemic control in adults with type 2 diabetes mellitus and to reduce the risk of major adverse cardiovascular events in adults with type 2 diabetes mellitus and established cardiovascular disease.

✔️ Pharmacology

Semaglo contains Semaglutide, a GLP-1 (Glucagon-like Peptide-1) analog. GLP-1 agonists like Semaglutide mimic the incretin hormone GLP-1, which plays several roles in glucose metabolism.

Mechanism of Action:

• Enhances Glucose-Dependent Insulin Secretion: By activating GLP-1 receptors on pancreatic beta cells, Semaglutide stimulates insulin secretion in response to elevated blood glucose levels.
• Suppresses Elevated Glucagon Secretion: Semaglutide reduces the secretion of glucagon from pancreatic alpha cells, which helps lower blood glucose levels by reducing hepatic glucose production.
• Slows Gastric Emptying: This delay in gastric emptying contributes to reduced postprandial glucose levels and aids in weight management by increasing satiety.

✔️ Dosage & Administration of Semaglo Subcutaneous Injection

Adult Dose

Indication: Type 2 Diabetes Mellitus

Semaglo is indicated as an adjunct to diet and exercise to improve glycemic control in adults with type 2 diabetes mellitus.

• Initial Dose: 0.25 mg subcutaneously once weekly for the first 4 weeks.
• Maintenance Dose:
  • After the initial 4 weeks, increase the dose to 0.5 mg once weekly.
  • If glycemic control is not achieved after at least 4 weeks on the 0.5 mg dose, the dose can be further increased to 1 mg once weekly.

Hepatic Impairment: No dosage adjustment is required.

Child Dose

• <18 years: Safety and efficacy have not been established for pediatric patients.

Renal Dose

• Renal Impairment: No dosage adjustment is required for patients with renal impairment.

Administration Information

Semaglo should be administered once weekly via subcutaneous injection in the abdomen, thigh, or upper arm. The starting dose is 0.25 mg, increased to 0.5 mg after 4 weeks, and can be further increased to 1 mg weekly if needed for glycemic control.

✔️ Interaction

• Semaglo may delay gastric emptying and impact the absorption rate of oral medications. Caution should be used when co-administering oral drugs that require rapid gastrointestinal absorption.

✔️ Contraindications

• Semaglo should not be used in patients with hypersensitivity to semaglutide or any of its excipients.

✔️ Pregnancy & Lactation

• Semaglo should not be used during pregnancy and should be discontinued at least 2 months before a planned pregnancy. It should not be used during breastfeeding due to potential risks to the child.

✔️ Precautions & Warnings

• Semaglo should not be used for type 1 diabetes mellitus or diabetic ketoacidosis. It should be discontinued if pancreatitis is suspected and not restarted if confirmed. Patients with diabetic retinopathy should be monitored closely.
• The safety and efficacy of Semaglo in children and adolescents below 18 years have not been established. No dose adjustment is required for elderly patients, nor for patients with mild to severe renal impairment or hepatic impairment. However, it is not recommended for patients with end-stage renal disease.

• Semaglo should be stored in a refrigerator at 2°C to 8°C. Do not freeze. Keep it out of reach of children.
