Tadalafil, found in Tada 20mg Tablet, is a phosphodiesterase type 5 (PDE 5) inhibitor used primarily for erectile dysfunction (impotence), benign prostatic hyperplasia (BPH), and sometimes pulmonary arterial hypertension (high lung blood pressure).

  • Erectile dysfunction: Tadalafil relaxes penile blood vessels, promoting blood flow when sexually aroused, and aiding in erections.
  • Benign prostatic hyperplasia (BPH): It relaxes bladder and prostate muscles, relieving BPH symptoms like urinary difficulties.
  • Pulmonary arterial hypertension: By relaxing chest blood vessels, it boosts lung blood flow and eases heart strain.

Take TADALAFIL as prescribed by your doctor, and its duration depends on your condition. Common side effects may include headache, back pain, muscle pain, leg and arm pain, nasal congestion, indigestion, and facial flushing. These usually resolve without medical attention.

Avoid TADALAFIL if you take nitrates (for angina), riociguat (for pulmonary hypertension), or alpha-blockers, have heart issues, low/high blood pressure, or if you're a woman. How to take Tada 20mg Tablet
  • Tada 20mg Tablet can be taken with or without food, following your doctor's instructions.
  • It should be taken at least 30-60 minutes before sexual activity when used for erectile dysfunction, and remember that sexual stimulation is necessary for it to work.
  • Regarding missed doses, Tada 20mg Tablet is typically used as needed, so missing a dose is less likely. However, if you're on a scheduled regimen and miss a dose, take it as soon as you remember. If your next dose is approaching, skip the missed one, and never double your dose to compensate for a missed one.
  • In the case of an overdose, do not exceed the prescribed dose. Seek immediate medical attention if you suspect an overdose. Swallow the tablet with a glass of water; do not crush or chew it. Your doctor will determine the appropriate dose and duration of therapy based on your age, body weight, and medical condition.

Indications:

Tadalafil is prescribed for Erectile Dysfunction (ED) and Benign Prostatic Hyperplasia (BPH). It works by inhibiting phosphodiesterase type 5 (PDE5), increasing cGMP levels, relaxing smooth muscles, and improving blood flow, resulting in penile erection. It also relaxes smooth muscles in the prostate and bladder, enhancing urinary flow.

Pharmacology

Tadalafil is a specific PDE5 inhibitor. It increases cGMP levels in smooth muscle cells, leading to smooth muscle relaxation and increased blood flow to the corpus cavernosum for penile erection. PDE5 is also found in smooth muscles of the prostate and bladder, and inhibiting PDE5 increases cGMP, relaxing these muscles, improving blood flow to the urinary tract, and widening the bladder neck for improved voiding.

Dosage & Administration of Tada 20mg Tablet

Adult Dose: For Erectile Dysfunction:

For Use as Needed: The typical starting dose for most patients is 10 mg taken before anticipated sexual activity. It can be increased to 20 mg or reduced to 5 mg, depending on how well it works and how well it's tolerated. The maximum recommended frequency of use is once per day.

For Once Daily Use: The starting dose for daily use is 2.5 mg, taken at approximately the same time each day, regardless of sexual activity. It can be increased to 5 mg based on individual response.

For Benign Prostatic Hyperplasia (BPH):

  • BPH: Take 5 mg orally once daily.
  • BPH with erectile dysfunction: Take 5 mg orally once daily, at approximately the same time each day, irrespective of sexual activity.
  • When using finasteride for BPH, take tadalafil 5 mg plus finasteride 5 mg orally once daily for up to 26 weeks.

For Pulmonary Arterial Hypertension:

  • Take 40 mg orally once daily. Avoid dividing the dose for more frequent administration.
  • For patients also taking ritonavir, start with 20 mg orally once daily for over a week, and it may be increased to 40 mg per day depending on tolerability.

For Hepatic Impairment (Erectile Dysfunction):

  • Mild-to-Moderate (Child-Pugh class A or B): Do not exceed 10 mg orally once daily.
  • Severe (Child-Pugh class C): Not recommended.

For Hepatic Impairment (Pulmonary Arterial Hypertension):

  • Mild-to-Moderate (Child-Pugh class A or B): Consider starting with 20 mg orally once daily.
  • Severe (Child-Pugh class C): Avoid use.

Renal Dose: For Erectile Dysfunction:

  • Mild (CrCl >51 mL/min): No dosage adjustment is needed.
  • Moderate (CrCl 30-50 mL/min): Do not exceed 5 mg orally once daily initially; the maximum dosage is 10 mg orally every 48 hours.
  • Severe (CrCl <30 mL/min and on hemodialysis): Do not exceed 5 mg orally every 72 hours.

For Pulmonary Arterial Hypertension:

  • Mild-to-Moderate (CrCl 31-80 mL/min): Start with 20 mg orally once daily and may be increased to 40 mg once daily based on tolerability.
  • Severe (CrCl <30 mL/min and on hemodialysis): Avoid use.

Tadalafil can be taken with or without food, and sexual stimulation is necessary for it to be effective. You can take it anywhere from 30 minutes to 12 hours before sexual activity, and its efficacy may last up to 24 hours after taking it.

Interaction

Tada 20mg Tablet can potentially interact with several drugs, including:

  • Anti-hypertensive drugs (amlodipine, lisinopril, losartan, metoprolol, riociguat)
  • Alpha-blockers (tamsulosin)
  • Anti-anginal drugs (nitroglycerin)
  • Anti-fungal drugs (ketoconazole, itraconazole)
  • Antibiotics (erythromycin, clarithromycin)
  • Anti-TB drugs (rifampicin)
  • Anticonvulsants (phenobarbital, phenytoin, carbamazepine)
  • Anti-HIV drugs (ritonavir)

There are no established drug-food interactions with TADALAFIL.

In terms of drug-disease interactions, it's important to inform your doctor if you have specific medical conditions, including:

Contraindications
