In some cases, yes, though this is usually to treat medical conditions other than ED. A person can take the medication daily or when needed, but they should first discuss dosage and frequency with their doctor.

Side Effects of Penfil 10mg Tablet

Common side effects may include headache, myalgia, respiratory tract infection, nasal pharyngitis, dyspepsia, flushing, back pain, and nausea. Less common side effects can involve changes in color vision, sudden vision loss, hearing loss, skin reactions like Stevens-Johnson Syndrome, angina, stroke, myocardial infarction, severe hypotension, and tachycardia.

Quick Suggestions:

• It is best to take it an hour before sexual intercourse. But, you can take it anytime between 30 minutes and 4 hours before sexual activity.
• Do not take it more than once a day.
• It may also be used for treating the urinary symptoms of an enlarged prostate gland (benign prostatic hyperplasia).
• It may cause dizziness or blurred vision. Do not drive or do anything requiring concentration until you know how it affects you.
• Avoid drinking alcohol as it can reduce your ability to get an erection and can make dizziness worse.
• Seek medical attention if the erection persists for more than 4 hours after sexual intercourse.
• Do not use Tadalafil if you have recently taken nitrates (medicines used in angina or chest pain), if you have had a heart attack in the past 3 months, or stroke or heart failure in the past 6 months.

Pharmacology

Tadalafil is a PDE5 (phosphodiesterase type 5) inhibitor. By inhibiting PDE5, it increases cGMP levels in smooth muscle cells. Elevated cGMP levels relax smooth muscles and enhance blood flow to the corpus cavernosum, facilitating penile erection. PDE5 is also present in the smooth muscles of the prostate and bladder wall. Inhibiting PDE5 raises cGMP concentrations, resulting in smooth muscle relaxation in the prostate and bladder. This relaxation may improve blood flow to the urinary system and enlarge the bladder neck opening, facilitating more efficient voiding.

Dosage & Administration of Penfil

Erectile Dysfunction: Typically, the recommended starting dose is 10 mg, with the option to increase to 20 mg or decrease to 5 mg based on individual needs. The maximum frequency of use is once daily, and the effect can last up to 36 hours.

Benign Prostatic Hyperplasia: The recommended dose is 5 mg taken at the same time daily.

Combined Erectile Dysfunction and Benign Prostatic Hyperplasia: The recommended dose is 5 mg daily.

Interaction

Tadalafil may interact with medications such as nitrates, alpha-adrenergic blockers, antihypertensives, alcohol, certain antacids, ketoconazole, ritonavir, erythromycin, itraconazole, grapefruit juice, other HIV protease inhibitors, rifampin, carbamazepine, phenytoin, and phenobarbital.

Contraindications

• Concurrent use of organic nitrates, nitrates, and nitric oxide donors.
• Men with certain cardiac conditions where sexual activity is not advised.
• Recent myocardial infarction (within 90 days) or stroke (within the last 6 months).
• Hypotension (blood pressure <90/50 mm Hg).
• Unstable angina, heart failure, uncontrolled arrhythmias, or hypertension.

Pregnancy & Lactation

Tadalafil is classified as pregnancy category B by the USFDA. Its use during pregnancy should only occur if the benefits outweigh the risks. There is limited information on Tadalafil excretion in human milk, so caution is necessary when administering it to breastfeeding women.

Precautions & Warnings

Caution is advised in patients with anatomical penile deformations, cardiovascular disease, history of myocardial infarction or stroke within the last 90 days, unstable angina, heart failure, uncontrolled arrhythmias, hypotension, uncontrolled hypertension, recent cerebrovascular accident, bleeding disorders, active peptic ulcer disease, liver disease, renal impairment, conditions predisposing to priapism, and concomitant use of CYP3A4 inhibitors.
