Treats diseases that cause hyperkinetic motor disorders (such as jerky, irregular, uncontrollable movements) associated with Huntington's chorea (an inherited neurodegenerative disease) in affected individuals

✔️ Pharmacology
  • Tetrabenazine reversibly inhibits human vesicular monoamine transporter type 2 (VMAT2), which leads to decreased uptake of monoamines (such as dopamine, serotonin, norepinephrine, and histamine) into synaptic vesicles.
  • This inhibition results in the depletion of monoamine stores from nerve terminals.
  • The mechanism is similar to reserpine but with less peripheral activity and is shorter-acting.
✔️ Dosage & Administration of Tetraben 12.5mg Tablet

Adult Dose:

For Huntington's Disease (chorea associated):

  • Start with 12.5 mg orally once daily.
  • After one week, increase to 12.5 mg every 12 hours.
  • Slowly titrate by weekly intervals of 12.5 mg/day to find a dose that reduces chorea and is well tolerated.
  • If the daily dose is between 37.5 to 50 mg, administer in divided doses every 8 hours.
  • For elderly patients, start with 12.5 mg daily and increase gradually.
  • If a dose higher than 50 mg/day is needed, test and genotype to determine metabolizer status. The maximum recommended daily dose is 100 mg/day or 37.5 mg per dose.

For moderate to severe tardive dyskinesia:

  • Start with 12.5 mg daily and increase gradually based on response.

Child Dose:

  • Safety and efficacy have not been established in children, so no specific dosing guidelines are available.

Special Populations:

  • Tetrabenazine is contraindicated in patients with hepatic impairment, as dose adjustment cannot ensure safe use.

Tetraben 12.5mg Tablet not be given with or within 14 days of discontinuation of MAOI therapy. It may block the action of reserpine and decrease the effects of levodopa. There is an increased risk of extrapyramidal side effects when given certain medications like amantadine, metoclopramide, and antipsychotics.
