✔️ Side Effects of

• Early Parkinson’s Disease: Common side effects (≥5% incidence) include nausea, dizziness, headache, insomnia, abnormal dreams, dry mouth, dyskinesia, anxiety, constipation, vomiting, and orthostatic hypotension.
• Advanced Parkinson’s Disease: Most commonly reported side effects include nausea and headache.

✔️ Dosage & Administration

• For patients new to levodopa therapy: The starting dose is 95 mg, taken three times daily for the first three days. On the fourth day, the dosage can be increased to 145 mg, and taken three times daily. Adjustments can be made based on patient response, with a maximum dose of 390 mg taken three times daily. Dosing frequency can be increased to a maximum of five times per day if necessary. The highest recommended daily dose is 2,450 mg.
• For patients switching from immediate-release carbidopa-levodopa: The doses are not directly interchangeable. The initial daily dose of Supadopa TR is determined based on the patient’s current levodopa intake, as outlined in the conversion table. Dosage adjustments should be made carefully to maintain symptom control while minimizing side effects like nausea and dyskinesia.

✔️ Drug Interactions

• Iron supplements and dopamine D2 antagonists (e.g., metoclopramide) may reduce the effectiveness of Supadopa TR.

✔️ Contraindications

• Not suitable for patients currently taking or who have recently (within two weeks) taken nonselective monoamine oxidase (MAO) inhibitors (e.g., phenelzine, tranylcypromine), as combining these can cause high blood pressure.

✔️ Pregnancy & Lactation

• Pregnancy: There is limited data on Supadopa TR’s effects in pregnant women. Animal studies indicate potential developmental risks, including teratogenic effects.
• Lactation: Levodopa has been found in human breast milk, and though carbidopa’s presence is unknown, levodopa may inhibit prolactin secretion, potentially affecting milk production.

✔️ Precautions & Warnings

• Patients should be monitored for excessive daytime sleepiness, withdrawal symptoms such as hyperpyrexia and confusion, cardiovascular complications, hallucinations, impulse control issues, dyskinesia, peptic ulcers, and glaucoma.
