✔️ Quick Suggestions:

• Take it shortly before or with the first main meal of the day (usually breakfast). Avoid skipping meals.
• Be careful while driving or operating machinery until you know how the Diamicron XR 60 Tablet affects you.
• It can cause hypoglycemia (low blood sugar level) when used with other antidiabetic medicines, alcohol, or if you delay or miss a meal.
• Always carry some sugary food or fruit juice with you in case you experience hypoglycemic symptoms such as cold sweats, cool pale skin, tremors, and anxiety.
• Your doctor may check your liver function regularly. Inform your doctor if you develop symptoms such as abdominal pain, loss of appetite, or yellowing of the eyes or skin (jaundice).

✔️ Indication

Gliclazide is a second-generation sulfonylurea drug with hypoglycaemic and potentially useful hematological properties. It stimulates the release of insulin from pancreatic β-cells by facilitating Ca+2 transport across the β-cell membranes and decreases hepatic glucose output.

✔️ Pharmacology

Gliclazide stimulates insulin secretion from pancreatic beta-cells, reduces insulin uptake, and glucose output by the liver, and increases insulin sensitivity at peripheral target sites. It decreases microthrombosis by partial inhibition of platelet aggregation and adhesion, and by restoring fibrinolysis with an increase in tissue plasminogen activator (t-PA) activity.

✔️ Dosage & Administration of Diatrol 80

Oral Type 2 Diabetes Mellitus (Adults):

• Initial Dose: 40-80 mg daily, gradually increased to 320 mg daily if necessary.
• Doses >160 mg daily: Given in 2 divided doses.
• Modified-Release Tablet: Initially, 30 mg once daily, may increase in increments of 30 mg up to a maximum of 120 mg daily. Interval between doses should be at least 1 month. For non-respondent patients, the dose may be increased after 2 weeks.

• Increased hypoglycaemic effect with phenylbutazone.
• Potentiation of blood glucose lowering effect with other antidiabetics (e.g. acarbose, insulins, metformin), beta-blockers, ACE inhibitors, H2-receptor antagonists, MAOIs, sulfonamides, clarithromycin, and NSAIDs.
• Increase in blood glucose levels with chlorpromazine, glucocorticoids, ritodrine, salbutamol, and terbutaline.
• Diminished hypoglycaemic effect with danazol.
• The increased anticoagulant effect of warfarin.
