Indications

Robic is an antibiotic that assists your body in fighting infections caused by bacteria and parasites. It is used to treat liver, stomach, intestine, vaginal, brain, heart, lung, and skin infections. Robic aids in the prevention of infection following surgery. It's also used to treat tooth infections, leg ulcers, and pressure sores. Treatment of susceptible anaerobic infections is among the conditions for which ornidazole is prescribed.

Pharmacology

Ornidazole, a 5-nitroimidazole derivative, is effective against protozoa and anaerobic bacteria. It is reduced to reduction products in susceptible organisms, which interact with DNA to destroy the helical DNA helix and strand, resulting in protein synthesis inhibition and cell death.

Dosage & Administration

Amoebiasis:

• Adults should take 500 mg twice a day for 5 days.
• Children: 10-25 mg per kg body weight divided into two doses.

Amoebic dysentery:

• Adults: 1.5 gm once daily for three days.
• Children: 40 mg/kg body weight, once daily for three days.

Giardiasis:

• Adults: Take 1.5 gm once a day for 1-2 days.
• For children, take 40 mg per kg body weight for two days.

Trichomoniasis: 

• For 5 days, take 1.5 gm once or 500 mg twice a day. The sexual partner should be treated concurrently.

Bacterial vaginosis: 

• One 500 mg tablet once daily for three to seven days, or three 500 mg tablets as a single dose.

Interaction

Ornidazole, like other imidazoles, has a minor risk of causing disulfiram-type responses. Due to a decrease in hepatic metabolism, using oral anticoagulants at the same time may increase the risk of bleeding. 5-fluorouracil clearance has been shown to be reduced by ornidazole.

Contraindications

Hypersensitivity to Ornidazole and other nitroimidazoles in the past. Ornidazole should not be used if you have a central nervous system problem, such as epilepsy or peripheral neuropathy.

Side Effects

• The most frequent side effects of ornidazole have been those that affect the gastrointestinal tract and central nervous system (vomiting, nausea, and epigastric discomfort) (dizziness, headache, and lassitude). Although this needs more research, ornidazole does not interact with alcohol in the same way that other nitroimidazoles do.
• Leukopenia has occasionally been reported during treatment. The most frequent adverse central nervous system (CNS) effects of ornidazole include headache, dizziness, lassitude or somnolence, fatigue, and weakness. The negative CNS effects of ornidazole could be less severe than those of metronidazole. Ornidazole has not been associated with any seizures in the investigations that have been conducted to date.

Pregnancy & Lactation

Clinical trials that are adequate have not been undertaken. Ornidazole should only be prescribed if the possible benefit outweighs the danger to the fetus or newborn.

Precautions & Warnings

• Ornidazole should be used with caution in patients who have ataxia, vertigo, or mental disorientation.
•  Blood dyscrasia, specifically mild leukopenia, has been described infrequently during long-term Ornidazole medication. 
• If leukopenia develops, the decision to stop treatment should be based on the severity of the infection.
