Atorvastatin is a selective inhibitor of the enzyme HMG-CoA reductase, which plays a key role in cholesterol synthesis by converting HMG-CoA to mevalonate. By blocking this enzyme, atorvastatin reduces cholesterol production in the liver and increases LDL receptor activity on hepatocyte surfaces, enhancing LDL uptake and clearance from the bloodstream.

Colostat is prescribed along with dietary control to help lower elevated levels of total cholesterol, LDL cholesterol, apolipoprotein B (Apo-B), and triglycerides when lifestyle measures alone are not sufficient.
It is used in:
Before starting therapy, patients should follow a cholesterol-lowering diet, which should be continued throughout treatment.
Adults (Primary hyperlipidemia / mixed dyslipidemia):
Usual starting dose is 10 mg once daily, which may be increased every 4 weeks up to a maximum of 80 mg daily depending on response.
Heterozygous familial hypercholesterolemia:
Start with 10 mg daily and titrate up to 40–80 mg daily as needed.
Pediatric patients (10–18 years):
Initial dose is 10 mg once daily; may be increased based on response, usually not exceeding 20 mg daily.
Cardiovascular risk reduction:
10–20 mg once daily, adjusted according to lipid response.
Atorvastatin may be taken at any time of the day, with or without food. Lipid levels should be reassessed after 2–4 weeks for dose adjustment.
Atorvastatin is contraindicated during pregnancy due to potential fetal harm. It should not be used in women who are pregnant, planning pregnancy, or suspected of being pregnant. Breastfeeding is also not recommended as the drug may pass into breast milk and potentially harm the infant.
Liver function should be monitored before and during treatment. Use caution in patients with alcohol use or liver disease. Therapy should be stopped if significant muscle pain or elevated CK levels occur.
Store in a cool, dry place away from light and moisture. Keep out of reach of children.
