✔️ Uses of Oragon 150

• Endometriosis.

✔️ Mode of Action

• Elagolix is an oral GnRH receptor antagonist.
• It inhibits GnRH signaling in the pituitary gland, reducing LH and FSH levels.
• This leads to decreased estradiol and progesterone, alleviating pain symptoms associated with endometriosis.

✔️ Dosage & Administration of Oragon 150

Before starting treatment:

• Ensure pregnancy is excluded.
• Start treatment within 7 days from the onset of menses.

Dosing Regimens:

Elagolix 150 mg:

• 1 tablet once daily, for up to 24 months.
• For moderate hepatic impairment, limit use to 6 months.

Elagolix 200 mg:

• 1 tablet twice daily, up to 6 months (especially in cases with coexisting dyspareunia).
• Do not exceed 6 months as it may reduce bone mineral density (BMD).

Hepatic Impairment:

• Severe hepatic impairment: Elagolix 200 mg is not recommended.

Children & Adolescents (<18 years):

• Safety and efficacy not established.

✔️ Side Effects of Oragon 150

• Hot flashes, night sweats, headache, nausea, amenorrhea, arthralgia, mood changes, bone loss, Insomnia, anxiety, elevated liver enzymes.

✔️ Overdose Effects of Oragon 150

• In case of overdose:
  • Monitor for adverse reactions and provide appropriate symptomatic treatment.

✔️ Administration Instructions

• Dosage Timing: Take at the same time daily, with or without food.

✔️ Interactions

With Medicines:

• May potentiate P-gp substrates (e.g., digoxin) and CYP2C19 substrates (e.g., omeprazole).
• May antagonize CYP3A substrates.
• Consider dose adjustment of oral midazolam and rosuvastatin.
• Reduced efficacy when used with CYP3A inducers or estrogen-containing contraceptives.

With Food:

• No known food interactions.

✔️ Contraindications

• Pregnancy: Risk of early pregnancy loss if exposed during pregnancy.
• Osteoporosis: Increases the risk of further bone loss.
• Severe hepatic impairment.
• Concomitant use with strong OATP1B1 inhibitors (e.g., cyclosporine, gemfibrozil).

✔️ Pregnancy & Lactation

Pregnancy:

• Contraindicated as it may increase the risk of early pregnancy loss.
• Discontinue immediately if pregnancy is confirmed during treatment.

Lactation:

• No data on the presence of Elagolix in human milk or its effects on breastfed infants.

✔️ Precautions & Warnings

Bone Loss:

• Long-term use can lead to irreversible bone density reduction. Assess BMD for women at risk.

Menstrual Irregularities:

• May reduce the ability to recognize pregnancy. Perform pregnancy tests if suspected.

Mood Changes:

• Monitor for suicidal ideation, depression, anxiety, or worsening mood disorders.

Liver Function:

• May cause dose-dependent elevation of liver enzymes. Monitor for signs of liver injury.

Contraception:

• Use non-hormonal contraception during treatment and for 1 week after discontinuation.
