• The most common side effects include CNS depression, such as drowsiness and ataxia.
• Behavior problems, abnormal eye movements, aphonia, tremors, vertigo, depression, amnesia, hallucinations, increased libido, insomnia, and palpitations may also occur.

• The addiction / habit-forming potential of this medicine is very high. Take it only as per the dose and duration advised by your doctor
• It may cause dizziness. Do not drive or do anything that requires mental focus until you know how this medicine affects you.
• Avoid consuming alcohol as it may increase dizziness and drowsiness.
• Inform your doctor if you are pregnant, planning to conceive or breastfeeding.
• Inform your doctor if you experience worsen anxiety, depression angry, or violent behavior and mania while taking this medicine.
• Do not stop taking medication suddenly without talking to your doctor as that may lead to nausea, anxiety, agitation, flu-like symptoms, sweating, tremor, and confusion.

✔️ Indication

• Lennox-Gastaut Syndrome (petit mal variant): Pandura 2 Tablet is indicated alone or as an adjunct in the treatment of this syndrome, which includes akinetic and myoclonic seizures.
• Absence Seizures (Petit Mal): It may be indicated in patients with absence seizures who have not responded to succinimides.
• Panic Disorder: It is indicated for the treatment of panic disorder, with or without agoraphobia. Panic disorder is characterized by unexpected panic attacks and associated worry about having more attacks.

✔️ Pharmacology

• Benzodiazepine Properties: Clonazepam, the active ingredient, has typical benzodiazepine properties, including anticonvulsant, sedative, muscle relaxant, and anti-anxiety effects.
• Mechanism of Action: It increases GABAergic neurotransmission in inhibitory synapses, leading to an increase in the effect of GABA, an inhibitory neurotransmitter, on the post-synaptic transmembrane chloride flux.
• Effect on Epilepsy: Clonazepam has been shown to quickly inhibit various types of paroxysmal activities in animal studies and human EEG studies, making it effective for generalized and focal epilepsy.

✔️ Dosage & Administration of

• Adults: The initial dose for seizure disorders should not exceed 1.5 mg/day, with incremental increases until seizures are controlled or side effects prevent further increases. The maximum daily dose is 20 mg.
• Panic Disorder in Adults: The initial dose is 0.25 mg given in two divided doses, with an increase to 1 mg/day for most patients after 3 days.
• Pediatric Patients: Dosing for infants and children up to 10 years old or 30 kg should be between 0.01 and 0.03 mg/kg/day, not to exceed 0.05 mg/kg/day, given in two or three divided doses.

✔️ Interaction

Clonazepam does not appear to affect the pharmacokinetics of phenytoin, carbamazepine, or phenobarbital. Its effects on other drug metabolisms have not been studied.

✔️ Contraindications

• Pandura 2 Tablet should not be used in patients with hypersensitivity to benzodiazepines or those with significant liver disease.
• It is contraindicated in acute narrow-angle glaucoma but may be used in open-angle glaucoma with appropriate therapy.

✔️ Pregnancy & Lactation

• Use in pregnancy should be considered if benefits outweigh potential risks. Breastfeeding is not recommended while using Clonazepam.
• Caution is advised when used in patients with multiple seizure disorders, and its concomitant use with valproic acid may lead to absence status.
