✔️ Pharmacology

• Tofacitinib is a partially reversible JAK inhibitor that prevents the body from responding to cytokine signals.
• It inhibits JAK, preventing STAT phosphorylation and activation, and works as an anti-inflammatory agent by inhibiting the JAK-STAT pathway.
• The JAK-STAT signaling pathway regulates cell transcription in hematopoietic and immune cell functions.

✔️ Dosage & Administration of Tofanib XR

• For rheumatoid arthritis: Tofacitinib 5 mg twice daily or Tofacitinib 11 mg once daily.
• For psoriatic arthritis: Tofacitinib 5 mg twice daily or Tofacitinib 11 mg once daily (in combination with nonbiologic DMARDs).
• For ulcerative colitis: Tofacitinib 10 mg twice daily for at least 8 weeks, followed by 5 or 10 mg twice daily.

✔️ Interaction

• Interaction with CYP3A4 inhibitors (e.g., ketoconazole) may require dose adjustment.
• Cautious use of medications causes CYP3A4 inhibition and CYP2C19 inhibition.
• Rifampin and other CYP inducers may reduce clinical response.
